黄芪多糖及丹酚酸对高糖诱导小鼠足细胞凋亡的影响

【摘要】 目的观察黄芪多糖与丹酚酸对高糖诱导的小鼠足细胞凋亡的影响。方法采用小鼠足细胞株,通过温度选择的培养方法诱导足细胞分化,分为正常对照组(5mmol/L)、甘露醇高渗对照组、高糖组、高糖+黄芪多糖组、高糖+丹酚酸组,分别于干预3h、6h、12h和24h后收集细胞;采用Annexin V-FITC和PI双染方法,用流式细胞仪观察各组足细胞的凋亡率。结果高糖组各时间点足细胞细胞凋亡率均高于同期正常对照组与高渗对照组(P<0.01);高糖+黄芪多糖组、高糖+丹酚酸组各时间点足细胞凋亡率均较高糖组降低(P<0.05,P<0.01);高糖+黄芪多糖组与高糖+丹酚酸组比较差异无统计学意义(P>0.05)。结论高糖可诱导足细胞凋亡,黄芪多糖与丹酚酸对高糖诱导的足细胞凋亡均有抑制作用。

【Abstract】 Objective To observe whether high glucose(HG) can induce apoptosis of podocyte and the protective effect of Astragalus polysaccharides(APS) and Salvianolic acid(SAL) on it. Methods Immortalized mouse podocytes were induced differentiation by temperature selection culture. The cells were treated with normal glucose (NG, 5μmol/L), mannitol(MA) control, HG(30μmol/L) and HG plus APS (0.2 g/L) or HG plus SAL(10 mg/L) for 3h,6h,12h and 24h. Cells were stained by Annexin V/PI and analyzed by flow cytometry. Results The rate of podocyte apoptosis in HG group was higher than those in NG and MA at each time point(P<0.01). Both of APS and SAL could restrain this trend(P<0.05, P<0.01), and there was no difference between APS L andSA(P>0.05). Conclusion HG can enhance podocyte apoptosis by suppressing APS and SAL.
