罗布麻提取物对神经元缺氧复氧损伤的保护作用
Protective effects of Apocynum venetum L.extract on rats’ cerebral cortical neurons from hypoxia/reoxygenation injury in vitro
【摘要】 目的研究罗布麻提取物(Apocynum venetumL.Extract,AVLE)对神经元缺氧复氧损伤的保护作用。方法将培养7天的新生SD大鼠皮层神经元细胞随机分为空白对照组、模型组和AVLE低、中、高浓度组。除空白对照组外,其余各组制作原代培养皮层神经元氧糖剥夺(OGD)再复氧模型,AVLE各浓度干预组分别加入浓度为50μg/ml、500μg/ml、5 000μg/ml的AVLE;分别观察各组OGD复氧后的0h、3h、6h、12h、24h、48h和72h等时间点细胞形态学、细胞毒作用、细胞线粒体活性、细胞凋亡率及凋亡相关蛋白表达量的变化情况。结果与模型组比较,AVLE能明显减少神经元LDH的释放,提高线粒体活性,对OGD神经元的保护呈一定的浓度与时间依赖关系。AVLE能显著抑制OGD神经元的凋亡,有效抑制Bax蛋白的表达,上调bcl-2的表达。结论 AVLE对OGD诱导的神经元损伤有一定的神经保护作用,可能成为新颖有效的防治急性缺血性卒中的神经保护药物之一。
【Abstract】 Objective To observe the neuroprotective effects of Apocynum venetum L.extract(AVLE) in an in vitro model of ischemia and its possible mechanisms involved.Methods Neuronal injury was induced by oxygen and glucose deprivation/reoxygenation(OGD/R),neuronal injury and viability,neuronal apoptosis,expression of apoptosis-related protein were analyzed by various techniques.Results AVLE significantly relieved neuronal injury,increased cell viability,and attenuated neuronal apoptosis induced by OGD.AVLE also depressed the overexpression of Bax and raised the level of bcl-2.Conclusion AVLE may become a novel and effective neuroprotective agent against cerebral ischemia.
