束缚应激致亚健康状态大鼠的行为学改变及中药的干预作用

【摘要】 目的 观察束缚应激致亚健康状态大鼠模型的行为学变化以及血清中CORT的含量,脑组织中TChE活性和5-HT含量变化,探讨中药的干预作用及其机制。方法 取雄性SD大鼠40只,体重320~360 g,随机分成4组,即正常对照组、模型对照组、抗衰老片组、六味地黄丸组,每组10只,采用束缚方法限制活动模拟人类心理应激因素,建立大鼠亚健康状态模型,在造模同时,用抗衰老片、六味地黄丸等中药进行干预,测定亚健康状态大鼠的耐力,学习记忆力及血清中CORT的含量,脑组织TChE活性和5-HT含量。结果 与正常对照组比,模型对照组大鼠的挣扎次数明显减少和静止时间明显延长(P<0.01),游泳路程和搜索平台的潜伏期明显延长(P<0.05);主动回避的潜伏期、电击次数和电击时间均有所延长,但差异不显著(P>0.05);血清CORT显著升高(P<0.05),脑组织TChE活性和5-HT含量明显升高(P<0.05)。而给予抗衰老片和六味地黄丸后,大鼠的挣扎次数明显增多,静止时间呈缩短趋势,其中抗衰老片组的挣扎次数差异显著(P<0.01);大鼠游泳路程显著缩短(P<0.05),且搜索平台的潜伏期有减少趋势(P>0.05),主动回避的潜伏期、电击次数和电击时间变化不明显。血清CORT含量、脑组织中TChE活性和5-HT含量显著降低(P<0.05, P<0.01)。结论 束缚应激致亚健康状态大鼠的的耐力和学习记忆力下降,血清CORT含量、脑组织中TChE活性和5-HT含量升高,而抗衰老片、六味地黄丸等中药可有效改善心理应激致亚健康状态大鼠的耐力及学习记忆能力,其机制可能与调节激素含量及神经递质功能有关。

【Abstract】 Objective To observe the changes of the behavior and serum CORT level, activity of TChE and content of 5-HT in brain tissue of sub-health state rat model induced by restraint stress, and to explore the effect of the Traditional Chinese Medicine and its mechanism. Methods Fourty male SD rats weighted 320-360 g were randomly divided into 4 groups: normal control group, model control group, Anti-ageing tablets group and Liuwei Dihuang pills group, 10 rats per group. The sub-health state rats were induced by restraint which simulating human physiological stress. At the same time, the rats in Anti-ageing tablets group and Liuwei Dihuang pills group were administrated by Anti-ageing tablets and Liuwei Dihuang pills. During the experiment, endurance capacity, behavioral memory ability,serum CORT level, activity of TChE and content of 5-HT in brain tissue of sub-health state rats were determined.Results Compared with normal control group, the struggle times of the rats in the model control group reduced obviously while the still time prolonged(P<0.01). The swimming distance and the latency period of searching for the plat remarkably prolonged(P<0.05); The latency period of active avoidance response,electric shock times and electric shock time had an increasing trend. Serum CORT level,activity of TChE and content of 5-HT in brain tissue of the rats obviously increased(P<0.05). After treatment of Anti-aging tablets and Liuwei Dihuang pills, the rats struggled more seriously, the still time presented a decreasing trend, especially the rat treated with Anti-ageing tablets(P<0.01); The swimming distance was decreased(P<0.05); the latency period of searching for the plat had a decreasing trend(P>0.05).The latency period of active avoidance response, electric shock times and electric shock time had no obvious difference; the serum CORT level, activity of TChE and content of 5-HT in brain tissue decreased remarkably(P<0.05, P<0.01). Conclusion The endurance capacity and the learning and memory ability of the sub-health state rat induced by restraint stress was decreased, the serum CORT level, activity of TChE and content of 5-HT in brain tissue was increased remarkably; Anti-ageing tablets and Liuwei Dihuang pills could improve the behavior of sub-health state rat, and the mechanism may be related to regulate hormone content and neurotransmitter function.
