重组表达SOD对野生小家鼠精子的冷冻保护作用
The Protective Effect of Recombinant SOD on Frozen Sperm from Mus musculus
【摘要】 目的 探讨重组表达的MnSOD或CuZnSOD对野生小家鼠精子冷冻损伤的保护作用。方法 以R18S3为基础冷冻保护剂,分别添加不同浓度重组表达的MnSOD或CuZnSOD,通过对各组ICR小鼠冻融精子的活力、质膜完整性、DNA损伤的比较,筛选出两种SOD的合理浓度,并将其添加于野生小家鼠的精子冷冻保护液中,观察添加SOD后,冻融野生小家鼠精子活力、质膜完整性、DNA损伤以及体外受精率的变化。结果 添加100U CuZnSOD和200U MnSOD后野生小家鼠冻融精子的活力以及质膜完整性都有显著性提高(P<0.05);添加200U MnSOD野生小家鼠冻融精子的体外受精率与对照组相比有显著提高(P<0.05),而添加100U CuZnSOD体外受精率则无显著变化。结论 在小鼠精子冷冻培养液中添加200U重组表达的MnSOD浓度,对野生小鼠精子冷冻损伤具有一定的保护作用。
【Abstract】 Objectctive To explore the protective effect of the recombinant MnSOD or CuZnSOD on the cryopreservation of wild mice sperm. Methods The R18S3 added with different concentrations of recombinant MnSOD or CuZnSOD was used as cryoprotectant to select two of the best concentration of SOD through comparison of the motility, membrane integrity, DNA damage of frozen-thawed ICR mice sperm in each group. Results The motility and membrane integrity of frozen-thawed wild mice sperm increased significantly(P<0.05) adding with 100U CuZnSOD and 200U MnSOD in R18S3compared to the control group. While there were no difference in the group of R18S3 with 100 U Cu Zn SOD adding, only in the group of R18S3 adding with 200U MnSOD the fertility rate of frozenthawed sperm significantly increased compared with the control group(P<0.05). Conclusion The cryoprotectant adding with 200 U recombinant MnSOD has a protective effect on the cryopreservation of wild mice sperm.
