猪繁殖与呼吸障碍综合征病毒Nsp2基因缺失株Jiangxi-3毒力的进一步研究

【摘要】 通过Reed-Muench法检测从江西发生持续高热的病猪体内分离的猪繁殖与呼吸综合征病毒(PRRSV)Nsp2基因缺失株Jiangxi-3第9代(Jiangxi-3F9)的病毒滴度;将此毒株接种于PRRSV和猪圆环病毒2型(PCV2)抗原与抗体均为阴性的60日龄健康猪进行猪半数致死量(LD50)的测定,并且对LD50试验中存活下来的试验猪再次进行较大剂量的攻毒试验,另将病毒接种于PRRSV和PCV2抗原和抗体均为阴性的6月龄健康猪,对所有攻毒试验猪通过体温检测、临床症状观察、RT-PCR、ELISA等方法进一步研究Jiangxi-3F9的毒力。结果表明:Jiangxi-3F9的病毒滴度为105.25TCID50·mL-1,Jiangxi-3F9病毒的LD50为102.·2mL-1。动物试验各项检测结果说明Jiangxi-3F9毒力强。

【Abstract】 The virulence of Nsp2 gene deletion Jiangxi-3 of PRRSV was further studied,which was isolated from pig of the so-called"Mystery Fever Disease Syndrome"in pig herds in Jiangxi Province in China.TCID50of the 9th generation of Jiangxi-3(Jiangxi-3 F9) was detected by Reed-Muench methond.The healthy pigs which were negative of antibody and antigen of PCV2 and PRRSV were divided into 3 groups for determining LD50,and the survival pigs were challenged again.The healthy 6-month-old pigs were challenged,which were negative of antibody and antigen of PCV2 and PRRSV.Virulence was evaluated by temperature detection,clinical symptom,RT-PCR and ELISA.The virus titer of the mutation 9th generation of Jiangxi-3 isolate were 105.25 TCID50·mL-1,and LD50 was 102.·2mL-1.The result showed the virulence of Jiangxi-3 F9 was intensified.
