大黄素对人肝癌细胞株SMMC-7721凋亡及CytC、Smac mRNA表达的影响

【摘要】 目的:研究大黄素对人肝癌细胞SMMC-7721的促凋亡作用及对CytC和Smac mRNA表达的影响。方法:人肝癌细胞SMMC-7721经大黄素处理后,用流式细胞术鉴定细胞凋亡,荧光定量PCR(Real time reverse transcription-polymerase chain reaction,Real time PCR)检测Cyt C和Smac mRNA的表达。结果:与对照组比较,流式细胞仪检测结果显示大黄素处理后的细胞早期凋亡率明显增高(P<0.05),实时荧光PCR检测结果显示大黄素处理后的细胞Cyt C和Smac mRNA表达与对照组比较明显增强(P<0.05)。结论:大黄素可通过促进线粒体促凋亡因子Cyt C和Smac mRNA的表达从而诱导肝癌细胞SMMC-7721凋亡,并可能通过线粒体通路、死亡受体通路及内质网通路等多条通路诱导肝癌细胞凋亡。

【Abstract】 Objective:To study the effect of emodin on apoptosis and the expression of Cyt C,Smac mRNA in hepatoma carcinoma cells.Methods:SMMC-7721 cells were treated with emodin,and the apoptosis rate was detected by Annexin V-FITC/PI double labeled flow cytometry.The expression of Cyt C and Smac mRNA were measured by Real time reverse transcription-polymerase chain reaction(Real time PCR).Results:Compared with that of the control group,the apoptotic rate was increased markedly(P<0.05).At the same time,the expression of Cyt C and Smac mRNA were increased significantly(P<0.05).Conclusion:Emodin can induce the apoptosis of SMMC-7721 cells by upregulating expression of Cyt C and Smac genes,and possibly induce apoptosis of hepatoma cells through mitochondrial pathway,death receptor pathway and endocytoplasmic reticulum pathway.
