非洛地平逆转代谢综合征大鼠心肌纤维化的作用
Mechanism of reversal of cardiac fibrosis in fructose-fed rats by Felodipine
【摘要】 目的探讨非洛地平逆转代谢综合征(MS)心肌纤维化的作用机制。方法雄性Wistar大鼠33只随机分为对照组(12只)和代谢综合征组(21只),采用10%高果糖饮水8个月建立MS动物模型,随机将9只代谢综合征组大鼠给予非洛地平。并采用苦味酸天狼猩红染色法检测大鼠左室心肌胶原含量,RT-PCR法检测心肌白细胞介素-18(IL-18)mRNA表达水平。结果与对照组相比,MS大鼠左室心肌组织胶原含量升高(P<0.01);IL-18mRNA表达水平升高(P<0.01);应用非洛地平干预治疗后,上述异常均减轻。结论非洛地平通过抑制IL-18表达在逆转MS心肌纤维化发生发展中起重要作用。
【Abstract】 Objective To investigate the mechanism of reversal of cardiac fibrosis in fructose-fed rats(FFRs) by Felodipine.Methods Thirty-three male Wistar rats were randomly divided into the control group(n=12) and the metabolic syndrome(MS) group(n=21).After the MS model was built by feeding with 10% fructose water for 8 months,9 FFRs were perfused into the stomach with Felodipine once daily.Then,myocardium tissues were collected.Collagen content was detected by Sirius Red staining and real-time RT-PCR was to detect mRNA expression of interleukin-18(IL-18).Results The collagen content of MS group was significantly higher than that of the control group(P<0.01).mRNA expression of IL-18 was significantly higher in the MS group than in the control group(P<0.01).After six-week treatment of Felodipine,the collagen content and IL-18 mRNA expression were significantly lower than those in the MS group.Conclusion Felodipine ameliorates cardiac fibrosis in MS via its suppressive effect on IL-18 expression.
