【摘要】 目的观察99Tc-MDP对佐剂性关节炎(AA)大鼠模型的影响,探讨99Tc-MDP在类风湿关节炎(RA)治疗中的抗炎作用及其机制。方法采用完全性弗氏佐剂足跖皮内注射法建立AA模型,随机将30只雄性Wistar大鼠分为正常对照组、AA模型组和99Tc-MDP治疗组,治疗组在造模10d后给予99Tc-MDP2.5mL/kg尾静脉注射治疗,1次/d,治疗15d;正常对照组和AA模型组给予相同体积的生理盐水尾静脉注射治疗,1次/d,治疗15d。观察大鼠左踝关节宽度、关节炎指数(AI),以及应用酶联免疫吸附法(ELISA)检测血清中的PGE1和PGE2的水平。结果①AA模型组、99Tc-MDP治疗组随着免疫时间的延长,左踝关节肿胀程度逐渐减轻,99Tc-MDP治疗组较AA模型组减轻明显(P<0.05);AA模型组和99Tc-MDP治疗组关节炎指数逐渐下降,99Tc-MDP治疗组较AA模型组下降明显(P<0.05);②各组血清中PGE1水平差异无统计学意义(P>0.05);AA模型组、99Tc-MDP治疗组血清中PGE2水平均高于正常对照组(P<0.05),AA模型组高于99Tc-MDP治疗组(P<0.05)。结论 99Tc-MDP可以降低AA大鼠左下肢踝关节宽度、关节炎指数及血清中的PGE2的水平,对抑制AA大鼠的炎症有一定疗效。更多还原

【Abstract】 Objective To observe the effects of 99Tc-MDP(technetium-99 conjugated with methylene diphosphonate) on adjuvant arthritis in rats,and study its anti-inflammatory mechanism.Method The AA(adjuvant arthritis) rat model was established by intra-dermal injection of complete Freud′s adjuvant.Thirty male Wistar rats were randomly divided into three groups: the control group,the adjuvant arthritis group and the 99Tc-MDP treatment group.In the treatment group,99Tc-MDP(2.5 mL/kg)was administered through the caudal vein on the 10th day and repeated 15 days later after arthritis was induced.The same volume of normal saline was given to the other two groups under the same protocol.Then,the left-right diameter of the left hind ankle,and arthritis index of each rat were recorded while PGE1 and PGE2 in peripheral blood were measured by ELISA.Results ①The left-right diameter of the left hind ankle in both the control group and the 99Tc-MDP treatment group was reduced with time,but the diameter in the 99Tc-MDP treatment group was more significantly reduced(P<0.05)than that of the AA group.The arthritis index of the AA group and the 99Tc-MDP treatment group both decreased with time,but the index decreased much more significantly(P<0.05)in the latter.②There were no significant differences among all groups on the level of PGE1(P>0.05),and the level of PGE2 of the AA group and the 99Tc-MDP treatment group were notably higher than the control(P<0.05).The PGE2 level of the control group was much higher than that of the 99Tc-MDP treatment group(P<0.05).Conclusion 99Tc-MDP can reduce the left-right diameter of the hind ankle and arthritis index in adjuvant arthritis rats.99Tc-MDP may have an anti-inflammatory effect on adjuvant arthritis rats.更多还原
