石榴皮水提物对大鼠离体结肠运动的影响及其机制的研究

【摘要】 观察分析石榴皮水提物对大鼠离体结肠段运动的作用及其途径。大鼠实验前禁食24 h,颈椎脱臼法处死,立即取靠近起始部的结肠段1.5 cm,浸在37℃恒温台氏液内。动物因给药不同(0.1,1.0,1.5,2.0,2.5mg/mL的石榴皮水提物,乙酰胆碱(ACh),ACh与1.0 mg/mL石榴皮水提物的混合液)被分成7个组。观察给药前、后10 min大鼠离体结肠段收缩幅度、收缩频率的变化。1.0、1.5、2.0 mg/mL的石榴皮水提物溶液对结肠段收缩频率较给药前均有明显的抑制作用(P<0.05),且呈一定的量-效正相关关系(P<0.05);1.0、2.0 mg/mL的石榴皮水提物溶液对结肠段收缩幅度较给药前均有明显的抑制作用(P<0.05);2.5 mg/mL的石榴皮水提物溶液引起的结肠段收缩频率(1.30±0.03次/min)较给药前(0.86±0.01次/min)有明显的加强作用(P<0.05),但对幅度的作用不明显(P<0.05);1.0mg/mL石榴皮水提物与ACh的混合液引起的结肠段收缩幅度与给药前比较,抑制率为71.00%,乙酰胆碱的抑制率为-48.60%,两者比较差异显著(P<0.05)。本实验结果提示1.0~2.0 mg/mL石榴皮水提物对离体大鼠结肠段收缩的幅度、频率均有抑制作用。这种抑制作用可能是通过调节ACh及其活动通路实现的。

【Abstract】 To determine the effect and pathway of water-soluble extraction from pomegranate peel on isolated colon segment in rat.The rats were fasted for 24 h before the experiment and then were sacrificed.Their colon segments(about 1.5 cm in length closing with cecum) were rapidly collected and immersed in Tyrode’s solution(37℃).The animals were divided into seven groups(0.1,0.5,1.0,1.5,2.0,2.5 mg/mL water-soluble extraction groups,ACh group and ACh+1.0 mg/mL water-soluble extraction group).The frequency and amplitude of the contractive waves of the isolated colon segment were recorded at least 10 min before and after drugs were administrated.The concentrations of 1.0,1.5 and 2.0 mg/mL water-soluble extraction from pomegranate peel significantly inhibited the frequency of isolated colon segment respectively(P<0.05),and showed dose-dependent relationship(P<0.05);The concentrations of 1.0,2.0mg/mL water-soluble extraction evidently decreased the amplitude(P<0.05);The concentration of 2.5 mg/mL water-soluble extraction significantly increased the frequency(P<0.05),but had no effect on amplitude(P<0.05);The admixture of 1.0 mg/mL water-soluble extraction and ACh significantly inhibited the contractive amplitude.The data suggest that the concentrations of 1.0~2.0 mg/mL watersoluble extraction from pomegranate peel inhibit the contractive frequency and amplitude of isolated colon segment significantly,and may be exert its action by regulating the ACh pathway.
