二甲亚砜对大鼠TBI中C-fos、C-jun基因表达干预作用的研究
The effect of dimethl sulfoxide on the expression of c-fos and c-jun gene in SD rat brain tissue after head injury.
【摘要】 目的研究二甲亚砜(DMSO)对SD大鼠弥漫性脑损伤(DAI)中c-fos、c-jun基因表达的干预作用,并探讨其机制。方法 60只SD大鼠随机分为空白组、对照组、实验组(DMSO干预组)。自制颅脑瞬间旋转模型(DAI模型)致伤,采用免疫组织化学技术检测脑损伤后c-fos、c-jun基因表达及变化规律。结果实验显示脑组织中c-fos、c-jun基因30min即有表达,1h有所增强,2h达到高峰,6h逐渐下降,且皮层、白质、脑干、丘脑、小脑均呈弥漫性表达。但实验组FOS蛋白和JUN蛋白的表达30min开始减少,1h、2h及6h明显少于对照组(P<0.05)。结论 c-fos、c-jun基因表达的变化规律和特点与颅脑损伤机制、继发性脑损伤有关。DMSO可使FOS蛋白和JUN蛋白的表达减少,2h其作用达到高峰。
【Abstract】 Objective To investigate the effect of dimethl sulfoxide on the expression of c-fos and c-jun gene in SD rat brain tissue after head injury.Methods 60 rats were divided into empty group、control and experimental group.Using the rat modle of diffuse axonal injury(DAI),the Fos and Jun positive neuros were observed in the cotrol and experimental group at 30min、1h、2h、6h after trauma by immunochemical method.Results Fos and Jun positive neurons could be find in cortex,white matter,brain stem,thalamus and cerebellum.Feebly Fos and Jun positive neurons were detected in the normal group,the Fos and Jun positive expression could be detected at 30min after DAI,increased at 1h after DAI,which reached manimal level at 2h,and began to decrease at 6h after DAI.The Fos and Jun positive expression of neurons in the control group was much stronger than that in the experimental group(P<0.05).Conclusion Expression of C-fos and C-jun gene may be related to different injury mechanism and secondary brain injury.The Fos and Jun positive expression in neurons could be reduced by dimethl sulfoxide,which reached manimal level at 2h.
