不同理化条件对GFP发光特性影响的研究

【摘要】 为了优化可用于保持绿色荧光蛋白(GFP)发光特性的理化条件,本文采用不同离心转速、温度、pH分别在不同作用时间下对表达GFP的HepG2细胞进行处理,然后在荧光酶标仪测定细胞荧光值。结果表明:转速从1000r/min提升到10000r/min,作用时间从5min到15min,GFP的发光度变化也停留在4个百分点之内;在-70℃~50℃这个范围内不会明显的破坏GFP的发光特性,但当温度达到90℃时GFP的发光明显减弱,且随着作用时间的延长发光越弱;当pH由2~12的变化过程中,GFP的发光度也由弱变强再变弱,时间越长对GFP的破坏越大,发光度值也就越小。

【Abstract】 In order to optimize proper physical chemistry condition that could maintain fluorescence of the green fluorescent protein (GFP),the present paper used the different efference rotation speed Temperature to fix GFP-expressing HepG2 cells at different time,and then GFP fluorescence was determined by microplate luminometer. The results showed that when the Speed raised from 1000r/min to 10000r/min,the action time from 5min to 15min,the change of the GFP luminosity is to stay within the 4 percentage points,when the Temperature change from 70 ℃ ~ 50 ℃,it is not to damage the optical properties of GFP significantly,but when the Temperature reach to 90 ℃ ,the luminescence of GFP is weaken。With the prolongation of the time ,the light is weaker. When the pH changes from 2 to 12,the luminosity of GFP is also changes from weak to strong and then weak. The longer of the time the greater of the destruction of GFP,the smaller the value of the luminosity.
