慢性髓性白血病和急性淋巴细胞白血病细胞中Wnt信号通路活性的差异
Activity of Wnt signalling in CML and ALL cells
【摘要】 目的:探讨慢性髓性白血病(CML)和急性淋巴细胞白血病(ALL)细胞中Wnt信号通路活性是否存在差异。方法:利用慢病毒基因转染法在经BCR-ABL融合基因诱导形成的CML和ALL细胞中建立Wnt信号通路报告系统BAT-RED,检测2种白血病细胞中Wnt信号通路活性;PCR方法检测2种细胞中BAT-RED基因整合情况;通过向培养细胞中添加氯化锂,人为增强Wnt信号活性,反向验证Wnt信号通路活性在2种细胞中存在差异。结果:PCR结果显示2种白血病细胞都稳定整合了Wnt信号通路报告基因BAT-RED,在42%(188/448)的慢性粒细胞白血病细胞中可以检测到Wnt信号通路被激活,而ALL细胞仅散在少数细胞中可以检测到Wnt信号通路报告基因BAT-RED被激活;添加氯化锂后,ALL细胞中Wnt信号通路活性增强。结论:CML细胞中Wnt通路活性明显高于ALL细胞,提示Wnt信号通路在2种类型白血病细胞的增殖和演进过程中具有不同作用。
【Abstract】 OBJECTIVE:To investigate whether Wnt signalling was differently utilized in chronic myelogenous leukemia(CML) versus acute lymphocytic leukemia(ALL) cells.METHODS: BCR-ABL fusion DNA fragment was introduced into mice bone marrow cells with intent to induce CML and ALL transformation,then Wnt reportor sysytem BAT-RED was successfully transfected into both CML and ALL cells with the lentiviral gene transduction method,and then the activity of Wnt signalling was evaluated in these two types of leukemia cells.The efficiency of BAT-RED integration was measured by PCR.The lithium choloride was added into culture medium to frocibly turn on the Wnt signalling in ALL cells,and to reversely verify the difference of Wnt signalling acitivy between CML and ALL cells.RESULTS: Although the integration of BAT-RED reportor gene was equal in CML and ALL cells,the activity of Wnt signalling pathway was significantly higher in CML cells by(42%(188/488)) than that in ALL cells.After lithium choloride was added to ALL cells,the Wnt reportor acitivity was enhanced.CONCLUSION: In CML cells,the activity of Wnt signalling pathway is higher than in ALL cells,which indicates that Wnt signalling plays different roles in CML and ALL,as for cell proliferation and tumor progression.
