芦荟对多柔比星外渗性损伤大鼠创面组织CD105和ICAM-1及VEGF影响的观察

【摘要】 目的:初步探讨芦荟促进大鼠多柔比星外渗性损伤创面愈合的机制。方法:将45只多柔比星外渗性损伤模型大鼠随机分为生理盐水组(1mL/kg)、硫酸镁组(1mL/kg)和芦荟组(2.5g/kg),每组15只,各组用药后第5天切除外渗性损伤部位的皮肤及皮下组织,间断缝合,术后第7天拆线,观察手术伤口Ⅰ期愈合情况和愈合时间。免疫组化法检测损伤部位的皮肤及皮下组织中的CD105、细胞黏附分子-1(ICAM-1)和血管内皮生长因子(VEGF)的表达情况。结果:三组大鼠的手术伤口Ⅰ期愈合率分别为33.3%、73.3%和86.7%,伤口愈合时间分别为(24.5±4.9)、(15.8±3.6)和(14.0±3.5)d,生理盐水组与另外两组之间,差异均有统计学意义,P<0.05。免疫组化结果显示,芦荟组与生理盐水组比较,VEGF、CD105的表达呈现上调,而ICAM-1的表达呈现下调。结论:芦荟能够显著促进外渗性伤口愈合,其作用机制可能与其促进VEGF、CD105的表达和抑制ICAM-1的表达有关。

【Abstract】 OBJECTIVE:To explore the possible mechanism of aloe gel on promote healing of injury in doxorubicin induced extravasation injury tissue in rats.METHODS:Forty five SD rats with extravasation injury model were randomly divided into NS group,magnesium sulfate group and aloe gel group.The exosmosis skin and subcutaneous tissues of rats were resected five days after treatments.Then the wounds were interruptedly sutured.When sutures were removed on the 7th day after operation,the condition of primary wound healing and healing time were recorded.Expressions of CD105,intercellular adhesion molecule (ICAM-1) and vascular endothelial growth facto (VEGF) in the exosmosis skin and subcutaneous tissues were detected by immunohistochemistry.RESULTS:The rates of primary wound healing were significantly higher in the aloe gel (86.7%) and magnesium sulfate (73.3%) groups than in the NS group (33.3%); while the healing time was significantly shorter (P<0.05) in the aloe gel 〔(14.0±3.5) d〕 and magnesium sulfate 〔(15.8±3.6) d〕 than that in the NS group 〔(24.5±4.9) d〕.Moreover,the expression level of CD105 and VEGF were higher in the aloe gel group than in the NS grouop.While the expression level of ICAM-1 was obviously lower in the Aloe gel group than NS group,The differencere were significant compared NS group.CONCLUSIONS:Aloe gel significantly reduced the extravasation injury and promote healing.The mechanism of the findings is associated with inhibit secretion of ICAM-1 and promote secretion of CD105 and VEGF in extravasation injury tissue.
