大肠埃希菌脂多糖脂质体的制备及对肿瘤的抑制作用

【摘要】 目的:制备临床分离的多重耐药大肠埃希菌TH1、TH2、TH6、TH7和TH10的脂多糖(LPS)脂质体并观察其对肿瘤的抑制作用。方法:采用逆向蒸发法制备大肠埃希菌LPS脂质体,通过电镜、粒径测定仪,鲎试验法(LAL)测定LPS脂质体的包封率。LPS脂质体免疫小鼠后将单肿瘤细胞悬液分别接种于小鼠腹腔和腓肠肌内,观察LPS脂质体对腹水瘤和实体肿瘤的作用。结果:大肠埃希菌LPS脂质体均为乳白色悬液,镜下可见LPS脂质体均呈规则的球形,粒径分布均匀。LAL结果推算包封率为99.0%~99.9%。TH1、TH2、TH6、TH7和TH10LPS脂质体免疫小鼠与对照组相比,腹水量和细胞总数明显减少且体质量增加,P<0.05,小鼠腓肠肌瘤体的体积均小于对照组,P<0.05。结论:所制备的大肠埃希菌LPS脂质体对肿瘤具有抑制作用。

【Abstract】 OBJECTIVE:To prepare liposomal lipopolysaccharide (LPS) from 5 strains TH1,TH2,TH6,TH7和TH10 of Escherichia coli(E.coli) isolated from clinic and observe its inhibitory effects on tumors.METHODS:Liposomal popolysaccharide of E.coli was prepared by the method of reverse evaporation.The properties were observed by a electron microscope,laser light scattering and limulus test.S180 tumor cells were inoculated in the abdominal cavity and gastrocnemius muscle of the mice immunized by the liposomal lipopolysaccharide and the effects were examined.RESULTS:The liposomal lipopolysaccharide was oyster white suspl in common temperature.It was multilayer round or oval saccule under the transmission electron microscope.The particle diameter was uniform.LAL test showed that entrapped efficiencies were 99.0%-99.9%.Tumor cells were inoculated to the mice immunized with lipopomal lipopolysaccharide of Escherichia coli,and ascites and total tumor cells of the immunized mice were less than those of the control group,and the body weights of the immunized mice were larger than those of the control group (P<0.05).Moreover,the volume of gastrocnemius muscle of the immunized mice was less than that of the control group (P<0.05).CONCLUSION:The prepared liposomal lipopolysaccharide from Escherichia coli has inhibitory effects on tumors.
