不同产地葫芦巴醇溶性化学成分的高效液相色谱-电喷雾质谱分析
Analysis of chemical components of Trigonella foenum-graecum L in different areas by high performance liquid chromatography-electrospray ionization-mass spectrometry
【摘要】 目的采用液质联用技术(LC-MS/MS)对不同产地中药葫芦巴中化学成分进行定性分析。方法运用DiamonsilTM C18(250mm×4.6mm,5μm)色谱柱,乙腈-冰乙酸-水梯度洗脱;质谱采用ESI离子源,正离子与负离子模式下采集数据。结果通过分析Q-TOFMS正、负离子质谱信息共检测到23个色谱峰,并结合对照品与相关文献数据其中鉴定出11个化合物结构,主要结构类型为黄酮类、皂苷类、生物碱类化合物。结论经高效液相色谱分离、质谱测定相对分子质量及相关文献数据信息检索可快速分析鉴定中药中的主要化学成分,为中药效应物质的阐明提供一定依据,为中药成分的分离鉴定提供快速准确的分析方法 。
【Abstract】 Objective To analyze and identify the chemical components of Trigonella foenum-graecum L of different producing area by LC-MS.Methods The analysis was performed on an Diamonsil C18(250mm×4.6mm,5μm)column.The mobile phases were acetonitrile and HAc and water in gradient elution.ESI source was used and data were acquired in positive and negative mode.Results The structures of 11 compounds of Trigonella foenum-graecum L were identified by direct comparison with MS of Q-TOF,the standard substances and the data of the literatures.Furthermore,the types of the compounds were flavones,saponins and alkaloids compounds.Conclusion It is a rapid and accurate method that the chemical compositions of traditional Chinese medicine can be identified in terms of the separation of high performance liquid chromatography,the accurate molecular weights measured by MS and other information,which can provide a method for active components research for TCM and TCM formulae.
