【摘要】 目的:检测内蒙古地区正常妊娠者、妊娠高血压(gestational hypertension,GH)病人HLA-DRB1等位基因频率;分析HLA-DRB1基因多态性与GH的相关性。方法:采用Luminex流式技术-序列特异性寡核苷酸探针反向杂交(flow cytometry-sequence specific oligonucleotide probe,FLOW-SSOP)方法对内蒙古地区80例正常妊娠者、40例GH病人进行HLA-DRB1基因分型,两组研究对象年龄、体重、孕次比较采用t检验。用直接计算法计算各等位基因频率和基因型频率。群体间等位基因频率比较应用χ2检验。结果:在受检的120例孕妇中共检出HLA-DRB1位点25个。两组孕妇HLA-DRB1等位基因频率表达的比较,GH组HLA-DRB1*0405检出10例,其基因频率为13.40%,而对照组仅6例,其基因频率为3.82%,两者比较,差异有显著性(P<0.05)。其它等位基因在两组间并未发现有显著性差异。结论:GH组HLA-DRB1*0405检出率高于正常妊娠组,内蒙古地区GH的发病与HLA-DRB1*0405等位基因呈正相关,DRB1*0405等位基因可能是GH的易感基因。更多还原

【Abstract】 Objective:To study the correlation between the polymorphism of HLA-DRB1 and susceptibility to gestational hypertension(GH) in Inner Mongolia population and further analyse the immunological mechanism of GH.Methods:The genotype of HLA-DRBI was examined by flow cytometry sequence specific oligonucleotide probes(FLOW-SSOP) in 40 GH and 80 healthy controls from Imer Mongdia.The age,body weight and gravidity of two groups were compared with t-test.The allelic frequencies of HLA-DRB1were calculated,in the group was identified by use of the χ2 test.Results:The result showed that 25 HLA-DRB1 alleles were found in 120 gravida in Inner Mongolia.The comparison of HLA-DRB1 allelic frequency:DRB1*0405 allele in the GH group was significantly higher than that in the healthy control group(13.40% vs3.82 %,χ2=6.563,P=0.01).No significant difference of other allelic frequencies was observed between the two groups.Conclusion:The HLA-DRB1*0405 allele frequency was higher in GH than those in the controls.This results suggest that HLA-DRB1*0405 might be the susceptible gene of GH in Inner Mongolia.更多还原
