TGF-β1对UVA照射人皮肤成纤维细胞MMP-1和MMP-3 mRNA
Effects of TGF-β1 on the expression of MMP-1 and MMP-3 mRNA in skin fibroblasts induced by UVA in vitro
【摘要】 目的:研究转化生长因子-β(1tansforming gowth fctor-beta1,TGF-β1)对长波紫外线(ultraviolet A,UVA)照射人皮肤成纤维细胞基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)和基质金属蛋白酶-3(matrix metalloproteinase-3,MMP-3)mRNA表达的影响,探讨TGF-β1对皮肤成纤维细胞的光保护机制。方法:通过体外培养人皮肤成纤维细胞,加入不同浓度TGF-β(10.1、1、10ng/ml)预处理细胞后UVA15J/cm2照射诱导细胞光老化,用四甲基偶氮唑蓝(MTT)法检测成纤维细胞增殖活性,半定量逆转录聚合酶链反应(RT-PCR)测定MMP-1、MMP-3mRNA表达。结果:UVA15J/cm2照射前给予不同剂量TGF-β1预处理,随TGF-β1剂量增加成纤维细胞增殖活性明显提高,MMP-1、MMP-3mRNA表达随着TGF-β1剂量增加而降低。结论:TGF-β1可提高UVA照射体外培养成纤维细胞增殖活性,减少UVA照射体外培养的皮肤成纤维细胞MMP-1、MMP-3mRNA表达,减少其对胶原的降解,对UVA照射皮肤成纤维细胞起保护作用。
【Abstract】 Objective To invesitgate the expression of matrix metalloproteinase-1 and matrix metalloproteinase-3 of skin fibroblasts induced by UVA radiation treated with various doses of TGF -β1, and investigate its photoaging protect mechanism in vitro. Methods The skin fibroblasts were cultured and irradiated with different doses of UVA. MTT assay was performed to analyze cell proliferation activity,and mRNA expression levels of MMP-1 and MMP-3 were detected by reverse transcription polymerase chain reaction (RT -PCR). Results The cell proliferation activity decreased with the UVA irradiation, but it increased when ading different doses of TGF-β1.The mRNA expression of MMP-1 and MMP-3 decrease with TGF-β1 increased,and it decreased in a dose-dependent manner. Conclusions TGF-β1 treatmented before UVA irradiation could improve skin fibroblasts proliferation activity in vitro. It is in a dosedependent manner to decrease the mRNA expression of MMP-1,MMP-3 on UVA irradiated fibroblasts. Which help protect skin fibroblasts from irradiation.
