【摘要】 目的探讨核转录因子kappaB(NF-κB)与大鼠肝纤维化形成的关系。方法用四氯化碳制造大鼠肝纤维化模型,以能够抑制NF-κB激活的N-乙酰-L-半胱氨酸(NAC)(100 mg/kg,ip)作为对照。在造模第4、8、12周末处死大鼠,动态观察大鼠肝脏活化NF-κB表达和活化肝星状细胞(HSCs)数量变化,并进行肝脏病理学检查。结果在各个造模时间点上,模型组大鼠肝脏炎症和纤维化评分明显高于正常大鼠(P<0.01),在第4、8周末肝脏枯否氏细胞(KCs)强烈表达活化NF-κB,肝脏炎症评分增加,NAC抑制NF-κB活化,肝脏炎症评分降低(P<0.01~0.05);随造模时间延长,表达活化NF-κB HSCs数量逐渐增多,活化HSCs亦增多,肝脏纤维化评分增加。NAC抑制NF-κB激活,活化HSCs减少,肝纤维化评分降低(P<0.01~0.05)。结论 NF-κB参与大鼠肝纤维化过程KCs和HSCs激活的调控。

【Abstract】 Objective To evaluate the role of NF-κB in rats liver fibrosis.MethodsLiver fibrosis in rats were induced by carbon tetrachloride(CCl4) subcutaneously,meanwhile,N-acetyl-L-cystein(NAC) at the dose of 100mg/kg body weight which could inhibit activity of NF-κB was treated by intraperitoneal.The rats were killed at the end of the 4th,8th,12th week,the expression of activated NF-κB in rats liver was analyed,the activated hepatic stellate cells(HSCs) in the liver specimen were counted,and histological examination of rats liver was measued.Results The score of hepatic inflammation and liver fibrosis in the model group increased significantly(P<0.01).At the end of the 4th and 8th week,Activated NF-κB was strongly expressed in Kupffer cells(KCs),the score of hepatic inflammation increased.When NAC inhibited the activation NF-κB,the score of hepatic inflammation decreased(P<0.01-0.05).At the end of the 12th week,the number of HSCs which expressed activated NF-κB were increased significantly,and the number of activated HSCs also increased,the score of liver fibrosis was increased obviously.With the inactivation of NF-κB by NAC,the number of activated HSCs decreased,and the score of liver fibrosis was also decreased(P<0.01-0.05).Conclusion NF-κB regulates the activation of KCs and HSCs.
