蛋白芯片技术在检测自身抗体中的应用探讨
The Approach and Application of Protein Chip Technology in the Field of Autoantibody Detection
【摘要】 目的评价蛋白芯片技术对自身抗体检测的敏感性和特异性,及其在常见自身免疫性疾病中的临床应用价值。方法采用蛋白芯片技术(在1张芯片上同时检测8项自身抗体),对214例自身免疫性疾病患者和43例健康体检者进行检测,并与免疫印迹法比较。结果检测抗Rib-P以蛋白芯片法的敏感性高于免疫印迹法(P<0.05),检测抗-SSB以蛋白芯片法的敏感性低于免疫印迹法(P<0.05),而抗-dsDNA、抗-Smith、抗-RNP、抗-SSA、抗Scl-70、抗J0-1的敏感性与免疫印迹法比较,差异无统计学意义(P>0.05),且这8项抗体的特异性均为100%。结论蛋白芯片法可同时检测8项自身抗体,操作方便、快速、敏感性高、特异性强,是常见自身免疫性疾病的辅助诊断方法 ,值得临床推广应用。
【Abstract】 Objective To evaluate the sensitivity and specificity of protein chip technique in the field of autoantibody,and it’s clinical value in common autoimmune diseases.Methods By applying protein chip technique,we detected eight autoantibodies on one chip at the same time,testing on 214 patients with autoimmune disease and 43 normal volunteers.Then,the outcome was compared with the data acquired by using immunoblotting technique(IBT).Results The sensitivity of protein chip technique was higher than that of immunoblotting technique when detecting anti Rib-P antibody(P<0.05),while the result was exactly reverse in the case of anti-SSB antibody(P<0.05).There was no difference between the sensitivity of protein chip technique and immunoblotting technique in detecting other antibodies,such as anti-dsDNA,anti-Smith,anti-RNP,anti-SSA,anti Scl-70 and anti J0-1 antibodies.Conclusion Protein chip technology is a fast and convenient technique with high sensitivity and specificity,which can be applied to detect eight autoantibodies at the same time.It is an auxiliary diagnostic method of common autoimmune diseases and worth popularizing in clinical application.
