【摘要】 目的探讨-80℃下不同冻存时间对脐血造血干细胞保存效果的影响。方法对-80℃下分别冻存1、2、3、4、8、16、32 w的脐血细胞进行复苏,比较各组间有核细胞数、CD34+数和台盼蓝拒染率的差异。结果脐血细胞在-80℃下保存1、2、3、4、8 w后,各组的有核细胞数和CD34+数的差异无统计学意义,而在冻存16、32 w后较1 w的差异有统计学意义(P<0.05)。台盼蓝拒染率在8、16、32 w较1 w的差异均有统计学意义(P<0.05),1、2、3、4 w各组间的差异无统计学意义。结论-80℃直接冻存对于短期(<8 w)脐血细胞的活性(有核细胞数、CD34+数和台盼蓝拒染率)无明显影响,但对于需要长期保存的脐血细胞的效果有影响。更多还原

【Abstract】 Objective To study the effect of different times of cryopreservation on hematopoietic stem cells under -80℃.Methods After the hematopoietic stem cells were frozen 1,2,3,4,8,16 and 32 w under-80℃,the number of nucleated cells and CD34+,trypan blue staining were compared.Results The differences of the number of nucleated cells and CD34+were not significant when the cells were frozen 1,2,3,4 and 8 w.Compared with the cells′activity at 1 w,the differences were significant at 16 and 32 w.Trypan blue staining showed that there were no significant distinctions at 1,2,3 and 4 w,but the differences were significant at 8,16 and 32 w.Conclusion There are no cryopreservation effects of short-term(<8 w) on hematopoietic stem cells′activities under-80℃,but long-term cryopreservation will influence the effects.更多还原
