重组人生长激素对体外培养的QGY-7701肝癌细胞生长的影响
Effects of rhGH on the growth of QGY-7701 human hepatic carcinoma cell lines in vitro
【摘要】 目的了解重组人生长激素(rhGH)对体外培养的QGY-7701肝癌细胞生长的影响。方法采用肿瘤细胞计数、噻唑蓝比色法了解7701肝癌细胞株在不同浓度rhGH(0、1、10、100、1000、10000ng/ml)作用下的药物敏感性,计算细胞生长率;用流式细胞计了解肝癌细胞在上述浓度rhGH作用下细胞周期各时相细胞比率、细胞增殖指数(PI)以及凋亡率的变化;采用放射配体法了解两种肝癌细胞生长激素受体(GHR)的表达情况。结果rhGH对QGY-7701肝癌细胞的生长无促进作用,各实验组在肿瘤细胞计数、MTT比色法、肝癌细胞DNA含量,细胞增殖指数的变化以及凋亡率等指标方面,与对照组同期比较,差异均无显著性;放射配体法未发现7701肝癌细胞表达GHR。结论rhGH对QGY-7701肝癌细胞的增殖无促进作用,原因可能与QGY-7701肝癌细胞不表达GHR有关。
【Abstract】 Objective To understand the effect of rhGH on the growth of QGY-7701 human hepatic carcinoma cell lines in vitro.Methods Tumor cell count and thiazole blue chromatometry were introduced to determine the sensibility of QGY-7701 to the different concentrations of rhGH(0、1、 10、100、1000、10000 ng /ml).Growth rate of tumor cells was calculated.The changes of cell proportion at each cell phase,cell proliferation index(PI)and apoptosis rate were calculated by flow cytometry.Finally,radioligand assay was used to detect the GHR expression of the hepatic carcinoma cell lines.Results To the QGY-7701 hepatic carcinoma cell line,each experimental group showed no significant differences on niether tumor cell count,thiazole blue chromatometry,DNA contents of the carcinoma cell,or the apoptosis rate compared with the control group.Radioligand assays showed no GHR expression was discovered in QGY-7701 hepatic carcinoma cell line.Conclusion rhGH has no effects on QGY-7701 hepatic carcinoma cell line in vitro because no GHR expression is discovered at all.
