高效毛细管电泳法分离4-羟基-苯异丙胺对映体
Separation of Enantiomers of 4-Hydroxy-Amphetamine by High Performance Capillary Electrophoresis
【摘要】 在内径100μm的毛细管内,以甲基丙烯酸甘油酯(GMA)和双(六-O-丁烯二酸单酯)-β-环糊精为单体原位聚合制得了手性毛细管电色谱整体柱,采用高效毛细管电泳法拆分了4-羟基-苯异丙胺对映体。考察了背景电解质的pH值、运行电压、柱温、流动相的配比等因素对分离度的影响。在背景电解质pH为4.2、运行电压20 kV、柱温18℃、乙腈-9 mmol.L-1磷酸盐缓冲溶液(60+40)为流动相、在4 min内成功地分离了4-羟基-苯异丙胺对映体,其分离度达到1.56。
【Abstract】 In-situ polymerization of monomers of glycerol methyl propenoiate(GMP) and bis-(6-O-monoester of butenedioic acid)-β-cyclodextrin was performed in a capillary of bore diameter of 100 μm to prepare the chiral capillary monolithic column,which was used for the HPCE separation of enantiomers of 4-hydroxy-amphetamine.Optimum conditions for the separation were found as follows: ① acidity of background electrolyte: pH 4.2;② temperature of capillary column: 18 ℃;③ separation voltage: 20 kV;④ mobile phase: mixture of acetonitrile and 9 mmol·L-1 PBS,mixed in the ratio of 60 to 40 by volume.Under the optimized conditions,the enantiomers of 4-hydroxy-amphetamine were satisfactorily separated within 4 min,having value of resolution factor 1.56.
