瘦素促进人肝癌细胞HepG2增殖的初步研究
Primary Study of Effect of Leptin on Proliferation of Human Hepatocellular Carcinoma Cells HepG2
【摘要】 目的探讨外源性瘦素对人肝癌细胞HepG2生长增殖的影响。方法体外培养HepG2,给予不同浓度瘦素(50ng/ml、100ng/ml、200ng/ml),MTT法观察瘦素对HepG2生长的影响,流式细胞仪检测瘦素对HepG2细胞周期的作用。结果 0ng/ml~200ng/ml瘦素在作用72h内呈现出剂量、时间依赖性地促进HepG2的增殖,不同浓度组之间及不同时间组之间,差异均具有统计学意义(P<0.01,P<0.01),并且随着瘦素浓度的增加,HepG2G0~G1期细胞比例逐渐下降(P<0.01),S期及G2-M期细胞比例逐渐上升(P<0.01,P<0.01)。结论瘦素在离体实验中具有明显的促进入肝癌细胞HepG2增殖的效应,此效应与促进细胞DNA合成、增强有丝分裂有关,提示瘦素有可能参与肝癌的发生、发展过程。
【Abstract】 Objective To investigate the effect of exogenous leptin on growth and proliferation of human hepatocellular carcinoma cells HepG2.Methods HepG2 cells were cultured in vitro and given leptin at different concentrations(50 ng/ml,100 ng/ml,200 ng/ml).To study the effect of exogenous leptin on the growth of HepG2 through MTT(2,5-diphenyltedtrazolium bromide)method.Meanwhile,cell cycles were measured by using the flow cytometry.Results Exogenous leptin(0 ng/ml-200 ng/ml)in 72 hours promoted the proliferation of HepG2 in a dose and time-dependent manner.There were significant statistical differences between groups of different concentrations and groups of different times(P<0.01,P<0.01).The cell ratio in G0-G1 phase decreased gradually(P<0.01)and that in S,G2-M phase increased gradually(P<0.01,P<0.01)as the concentration of leptin increased.Conclusion Leptin could obviously promote proliferation of human hepatocellular carcinoma cells HepG2 in vitro,which could be related with stimulation of DNA synthesis and enhancement of mitotic activity,and leptin maybe play a role in genesis and development of hepatocellular carcinoma.
