抗结核天然药物筛选的巨噬细胞模型研究

【摘要】 目的通过构建小鼠腹腔巨噬细胞结核杆菌感染模型,小鼠抗结核含药血清制备体系,建立抗结核药物筛选的巨噬细胞模型评价体系.方法按常规方法取小鼠腹腔巨噬细胞通过一定比例感染结核杆菌,建立小鼠腹腔巨噬细胞感染模型.利用已知及新研发的抗结核中西药物制备小鼠含药血清,与小鼠巨噬细胞结核杆菌感染模型共培养,破胞溶解后转种罗氏培养基,通过生长情况对比,建立巨噬细胞结核杆菌感染模型抗结核药物筛选的评价体系.结果成功构建小鼠腹腔巨噬细胞结核杆菌感染模型,利用该模型对利福平、猫爪草、欣痨宁等抗结核中西药物的含药血清进行抗结核效应评价,与现有资料报道相一致.结论巨噬细胞结核杆菌感染模型对抗结核药物筛选的评价体系可用于各种抗结核药物的筛选,弥补了抗结核药物筛选的体外MIC法及体内抗结核活性研究的不足.

【Abstract】 Objective To construct tubercular model of peritoneal macrophages and preparation system of serum containing anti-TB drug in mice,and establish evaluation system of the macrophage model for screening anti-TB drug.Methods We collected peritoneal macrophages of mice by conventional methods and infected them with Mycobacterium tuberculosis to establish peritoneal macrophage model for screening anti-TB drugs.We established preparation system of serum containing anti-TB drug from mice,and co-cultured serum containing anti-TB drugs with macrophage model infected by Mycobacterium tuberculosis,and then dultured the destroyed macrophage in the Lowenstein-Jensen medium,we established the evaluation system of the macrophage model infected by Mycobacterium tuberculosis for screening anti-TB drug by comparing the growth.Results We successfully constructed the macrophage model infected by Mycobacterium tuberculosis and made use of this model to evaluate respectively the anti-TB drug effect of serum containing the RFP,Maozhuacao,Xinlaoning.The results were similar with the existing data.Conclusion The evaluation system of the macrophage model infected by Mycobacterium tuberculosis for screening anti-TB drug can be used for screening various anti-TB drugs and make up the limitations of the MIC in vitro and the study of anti-TB activity in vivo.
