HEK293、COS7、7721、HepG2和PC12细胞内神经鞘磷脂合成酶活性比较

【摘要】 目的:分析比较HEK293、COS7、7721、HepG2和PC12的细胞内神经鞘磷脂合成酶(SMS)的活性。方法:收集处于对数生长期的HEK293、COS7、7721、HepG2和PC12细胞,每种细胞的细胞浓度相同;5种细胞均建立2个酶促反应体系,反应时间分别是2 h和3.5 h;薄层层析和灰度扫描分析比较SMS活性强弱。结果:HEK293细胞内的SMS活性较强。结论:建立了有效检测不同来源细胞SMS活性比较的方法,发现HEK293细胞具有较强的SMS活性,可以更好的用于神经鞘磷脂代谢的研究。

【Abstract】 Objective: To analyze and compare the activities in HEK293,COS7,7721,HepG2 and PC12 sphingomyelin synthase(SMS) which were commonly used in medical research.Methods: The HEK293、COS7、7721、HepG2 and PC12 cells was collected in each logarithmic phase,the concentration of which was the same.2 zymo-exciter systems was established by 5 kinds of cells.The activities of SMS were tested and compared at 2h and 3.5h.The SMS activities were analyzed and compared for their strength and weakness with thin layer chromatographic(TLC).Results: The SMS activity of HEK293 is the highest in HEK293 cells.Conclusion: In this study,an effective method was established to compare the activities of sphingomyelin synthase in different cell lines.The HEK293 cells has stronger SMS activity which means it can be better used in sphingomyelin research.
