冻干羊膜致Ⅰ型超敏反应的变应原性研究

【摘要】 目的研究冻干人羊膜的变应原性及其致敏后发生Ⅰ型超敏反应的可能性。方法建立豚鼠全身主动过敏实验动物模型。将动物随机数字方法分为冻干羊膜组、新鲜蛋清阳性对照组和PBS液阴性对照组,每组20只。观察豚鼠在致敏期和激发后的Ⅰ型超敏反应,化学荧光法分析外周血组胺含量,血液流变分析系统检测全血高切变率黏度、全血低切变率黏度、血浆黏度、红细胞聚集指数4项血液流变学指标。结果致敏期间各组豚鼠的体质量变化差异无统计学意义(P>0.05);激发后冻干羊膜组豚鼠与阴性对照组均无异常反应;冻干羊膜组外周血组胺含量及4项血液流变学指标均与阴性对照组差异无统计学意义(P>0.05),与阳性对照差异有统计学意义(P<0.05)。结论冻干羊膜一般不具有变应原性,不会引起Ⅰ型超敏反应。

【Abstract】 Objective To investigate the allergenicity of freeze-dried amniotic membrane(FDAM) and the possibility of occurrence of type Ⅰ hypersensitivity reactions after sensitization.Methods In systemic active anaphylaxis model,60 guinea pigs were randomly divided into three groups:FDAM group,albumen group(positive control) and PBS group(negative control),with 20 guinea pigs in each group.Blood samples were obtained after allergen challenge.The concentrations of blood histamine were determined by chemical fluorescence analysis.Whole blood viscosity at high and low shear rates,plasma viscosity,and erythrocyte aggregation index were tested by hemorheology analysis system.Results There were no significant differences in body weight change among the groups(P>0.05).No abnormal reaction was found in FDAM and PBS groups after sensitization.Blood histamine concentrations and hemorheologic markers in FDAM group were significantly lower than those in positive control group(P>0.05),but obviously different from those in negative control group(P<0.05).Conclusion FDAM does not have the allergenicity and can not cause type I hypersensitivity reactions.
