银杏酸的分离、鉴定及对5种蔬菜病原真菌的抑制作用

【摘要】 银杏外种皮采用体积分数为70%乙醇回流提取得到含活性成分银杏酸的粗提物,用乙醚、质量分数为5%Na2CO3、质量分数为5%的NaOH萃取分离,经硅胶柱层析纯化得银杏酸同系物;采用HPLC进行定量、纯度测定,用LC/MS对其中的银杏酸进行定性鉴定。实验测得银杏外种皮中银杏酸含量为3.12%;银杏酸同系物纯度为86.7%;平均回收率为97.3%,RSD为1.3%(n=5)。HPLC和LC/MS图谱表明该方法获得的银杏酸同系物无其他杂质峰,纯度高。银杏酸对甘蓝黑斑病菌抑制作用较强,EC50为24.80 mg/mL。

【Abstract】 The crude extract of active ginkgolic acids in sarcotesta of ginkgo biloba was gained with 70% alcohol circumfluence distillation,then it was extracted with other,5% Na2CO3,5% NaOH;the homologue of ginkgolic acids was purified by silica gel column chromatography.In this study,the quantity and purity of ginkgolic acids were determined by HPLC,identified by LC/MS.The results showed that the content of ginkgolic acids in sarcotesta of ginkgo biloba was 3.12%;the average recovery was 97.3%,RSD was 1.3%(n=5);the purity of ginkgolic acids was 86.7%.HPLC and LC/MS patterns showed that the ginkgolic acids obtained with this method had no impurity peak,their purity were high.Bioassay to the ginkgolic acids for the tested fungi showed that the ginkgolic acids were effective to Alternaria brassicae,in which the EC50 was 24.80 mg/mL.
