胰岛素促进体外兔角膜缘干细胞的增殖
Insulin promotes the proliferation of rabbit limbal stem cells in vitro
【摘要】 目的从原代培养获得角膜缘干细胞(LSCs),探讨不同浓度胰岛素对角膜缘干细胞增殖作用的影响。方法取兔角膜组织体外培养获得角膜缘干细胞,并行免疫组织化学及透射电镜鉴定。5mg/L、10mg/L、20mg/L、40mg/L、80mg/L及100mg/L胰岛素作用后,以四甲基偶氮唑盐法检测胰岛素对角膜缘干细胞增殖能力的影响。结果 5mg/L和10mg/L浓度的胰岛素有促进角膜缘干细胞增殖的作用,与对照组比较有显著性差异(P<0.05);5mg/L与10mg/L组比较也有显著差异(P<0.05);20~100mg/L组与对照组比较无显著性差异(P>0.05)。结论胰岛素促进培养角膜缘干细胞的增殖,最适宜浓度为5mg/L。
【Abstract】 Objective To get limbal stem cells(LSCs)by primary culture and explore the influence of insulin on LSCs’proliferation.Methods The cells from the rabbit corneal limbal tissue were cultured and identified by immunohistochemical staining and transmission electron microscopy.The LSCs were treated with 5mg/L,10mg/L, 20mg/L,40mg/L,80mg/L and 100mg/L insulin respectively.Then methyl thiazolyl tetrazolium(MTT)assay was used to explore the influence of insulin on LSCs’proliferation.Results The experiment groups at the concentration of 5mg/ L and 10mg/ L had significantly higher cell viability than that of the control group(P<0.05),and so were the 5mg/L group and 10mg/L group ( P<0.05 ).But there were no significant difference between the experimental groups at the concentration of 20mg/ L,40mg/ L,80mg/ L,100mg/ L and the control group ( P>0.05).Conclusion Insulin enhances the proliferation of the LSCs in vitro effectively.The optimal concentration is 5mg/L.
