F9细胞的培养和全细胞抽提物获取方法的比较

【摘要】 目的对F9小鼠畸胎瘤细胞进行培养,探索理想的获取全细胞抽提物的方法。方法应用蛋白质抽提试剂盒、反复冻融循环和超声波裂解法进行F9全细胞抽提物的获取。结果通过蛋白质抽提试剂盒和反复细胞冻融循环的方法所获取的细胞抽提物渗透压和浓度偏低,而选用超声波裂解法所获取的细胞抽提物,无论从pH,渗透压,还是蛋白浓度方面分析均与理想值接近。结论选择超声波裂解法作为获取F9小鼠畸胎瘤全细胞抽提物的理想方法 。

【Abstract】 Objective To culture the F9 mouse teratoma cells and find the perfect method for acquiring cellular extract. Methods The cellular extract of the F9 mouse teratoma cells was performed by protein extract kit, freeze-thaw cycle and ultrasonic methods respectively. Results The osmotic pressure and protein concentration in cellular extract of the F9 mouse teratoma cells were much lower by means of protein extract kit or freeze-thaw cycle than by ultrasonic method. The pH value, osmotic pressure and concentration of protein acquired with ultrasonic method were similar to the ideal level. Conclusion Ultrasonic method was the perfect method for acquiring F9 cellular extract compared with protein extract kit or freeze-thaw cycle method.
