小胶质细胞的活化对其Aβ吞噬能力的影响
The effect of microglia cells activation on their phagocytic ability of β-amyloid protein
【摘要】 目的研究活化的小胶质细胞对β淀粉样蛋白吞噬能力的影响。方法以Aβ1-42直接作用于小胶质细胞系细胞BV-2,NO检测试剂盒检测培养液中NO含量;CyQuent增殖试剂盒检测小胶质细胞BV-2的增殖情况,以及共聚焦显微镜观察小胶质细胞BV-2的Aβ吞噬能力。结果在Aβ1-42作用下,小胶质细胞BV-2释放的NO增加,小胶质细胞BV-2增殖能力增强,并且小胶质细胞BV-2的Aβ吞噬能力增强。结论Aβ沉积诱导小胶质细胞BV-2NO释放和细胞增殖,从而使其从静止态转为活化状态,最终导致活化的小胶质细胞Aβ吞噬能力增强。
【Abstract】 Objective To study the phagocytic capacity of active microglia cells to β-amyloid protein(Aβ).Methods Aβ1-42 was directly cocultured with microglia cells line BV-2,NO detection kit was used to detect NO content in culture medium,the CyQuent proliferation detection kit to detect BV-2 microglia cell proliferation,as well as confocal microscope to show phagocytosis of microglia BV-2 cells to Aβ.Results After Aβ1-42 was added,NO release of microglia BV-2 cells increased,microglia BV-2 cells proliferation increased,and the phagocytosis of Aβ by microglia BV2 cells increased compared with control.Conclusion Aβ deposition-induced NO release and cell proliferation of microglia BV-2 cells enhanced the phagocytic capacity of Aβ by active microglia cells.
