王氏保赤丸对抗生素诱导肠道菌群失调小鼠肠道保护作用及肝脏毒性的影响研究

【摘要】 目的:通过观察王氏保赤丸对小鼠肠道组织,肝脏组织的影响,探讨该药是否既能清热泻下又能避免苦寒伤胃,以及对肝脏的毒性作用,为合理使用该药提供实验依据。方法:建立抗生素引起菌群失调小鼠模型和不同剂量的王氏保赤丸灌胃小鼠模型,与正常小鼠比较,评价王氏保赤丸对小鼠肠道组织及肝脏的影响情况。结果:灌胃14 d后王氏保赤丸高、中、低剂量组均未出现模型组的小肠绒毛充血、变平、变短、萎缩、倒伏等现象。与模型组对比,高、中、低剂量组肠黏膜均较完整,中、高组黏膜固有层见较多炎性细胞浸润,但小肠绒毛均有变长、杯状细胞增多;回盲部的病理结果提示该药高、中、低剂量组对回盲部均无损伤。肝脏病理结果显示王氏保赤丸高、中、剂量组对肝细胞均有损害作用,低剂量组对肝细胞损害较轻微。结论:经合理配伍之后,该药保存了大黄、黄连等苦寒药物的清热解毒泻下作用,对肠道黏膜不仅无损伤,且有一定的保护作用。该药对肝脏有损伤,有一定的肝毒性,肝脏损伤者应慎用。

【Abstract】 Objective To observe the change of Wanshibaochiwan (WSBCW) in the intestine and liver of antibiotic-induced intestinal dysbacteriosis mice,and provide data and evidence for proper use of it in clinic.Method Antibiotic-induced intestinal dysbacteriosis mice model group and WSBCW groups of various doses were made to compare the morphology of their intestine and liver between groups and with normal mice.The mice were given gastric lavage for14 days by high dosage,middle dosage and lowdosage WSBCW[147 mg/(kg.d),49 mg/(kg.d),17 mg/(kg.d)] in WSBCW groups,and Lincomycin(0.4 ml/d) in model group,NS(0.4 ml/d) in normal group.The structure of appendix and small intestine,liver were examined after 14 days.Results The congestion,flattening,shortening,atrophy,and lodging of small intestinal villosity microvessels of mice that showed in model group were not showed in 14-day WSBCW groups.The intestinal mucosa were relatively complete in all doses WSBCW groups compared to model group.There were more inflammatory infiltration in mucoderm in high and medium WSBCW groups,but with lengthening of small intestinal villosity microvessels,and increasing of goblet cells.Pathological observation showed that all doses groups of WSBCW did not damage ileocecal junction.Liver cell damages were found in medium and high dose WSBCW groups,but the damage was very slight in low dose group.ConclusionWSBCW is safe and even shows protective effect to intestinal tract,but shows hepatotoxicity,patients with liver injury should be cautious when using it.
