激光扫描共聚焦显微镜在骨髓基质细胞移植中的应用
Application on transplantation of bone marrow stromal cells with Laser scanning confocal microscope
【摘要】 目的:探讨激光扫描共聚焦显微镜(laser scanning confocal microscope,LSCM)技术在骨髓基质细胞(Bone Marrow Stromal Cells,BMSCs)移植中的应用。方法:使用活细胞示踪剂CM-DiI、细胞核探针DAPI和DNA合成前体物BrdU体外标记BMSCs,移植到动物模型体内,利用LSCM技术,观察BMSCs在受体组织内的分布和分化。结果:体外标记BMSCs后再移植到受体,使用LSCM技术可以观察到BMSCs定位于受体心肌组织和脑组织,并有部分BMSCs表达相应的组织特异性蛋白Cx43和GFAP。结论:利用LSCM技术观察BMSCs在受体组织内分布和分化是一种简单可行的方法。
【Abstract】 Objective:Location and differentiation of bone marrow stromal cells(BMSCs) were investigated with Laser scanning confocal microscope(LSCM).Methods:After BMSCs in pig and rats were loaded with Cell-Tracker CM-DiI,DAPI and BrdU, they were transplantated into animal model,respectively.Location and differentiation of BMSCs in recipitent was observed with LSCM.Result:Location of BMSCs in recipient tissues was detected.Furthermore,BMSCs could differentiate into myocardial cell or glial cells with expression of Cx43 or GFAP in every group.Conclusion:It is a simple and effective method to observe the Location and differentiation of BMSCs with LSCM.
