针灸治疗帕金森病可能氧化应激作用机制的研究
The possible mechanism of oxidative stress on acupuncture treatment in the rat of parkinson’s disease
【摘要】 目的探讨针灸对治疗帕金森病(PD)可能的氧化应激作用机制。方法在脑定位仪下,将6-羟基多巴注入左侧纹状体造模,连续2周针灸治疗,1/d。2周后,测定血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)含量。结果与正常组比较,大鼠PD造模成功后,SOD活力升高(104.33±7.91U/μgvs112.85±3.33U/μg,P<0.05),GSH-PX活力降低(709.65±29.86U/μg-1·min-1vs686.94±21.94U/μg-1·min-1,P<0.05),MDA含量升高(22.93±1.85nmol/mgvs28.74±0.79nmol/mg,P<0.05)。与模型组比较,针灸组、美多巴+针灸组均能提高SOD活力分别为130.14±5.08、124.55±12.40U/μg,(P<0.01),提高GSH-PX活力分别为777.55±39.36、700.51±18.93U/μg-1·min-1,(P<0.01,P<0.05),降低MDA含量分别为15.35±1.68、18.03±3.31nmol/mg,(P<0.01),且针灸组作用显著。结论在6-羟基多巴毁损纹状体模型中,针灸治疗显著提高SOD活力和GSH-PX活力,降低MDA含量,针灸可能参与PD氧化应激机制的调节。
【Abstract】 Objective We investigated the possible neuroprotective effect of acupuncture on the mechanism of oxidative stress in the rat of Parkinson’s disease.Methods The rats received a unilateral 6-hydroxydopamine(6-OHDA)injection into the left striatum.Acupuncture intervention was performed within 2 weeks by post-lesioning.The expression of SOD、GSH-PX and MDA was evaluated by chemistry luminesce method、DTNB colorimetry and TBA reaction method.Results Compared with normal group,the effect of SOD is increased in model group(104.33±7.91 U/μg vs 112.85.±3.33 U/μg,P<0.05),the effect of GSH-PX reduced(709.65±29.86 U/μg-1·min-1 vs 686.94.±21.94 U/μg-1·min-1,P<0.05),The content of MDA increased(22.93±1.85 nmol/mg vs 28.74.±0.79 nmol/mg,P<0.05).Compared with model group,acupuncture group & Madopa/acupuncture group can increase the effect of SOD(130.14±5.08、124.55±12.40 U/μg,P<0.01)and GSH-PX(777.55±39.36、700.51±18.93 U/μg-1·min-1,P<0.01,P< 0.05),decrease the content of MDA(15.35±1.68、18.03±3.31 nmol/mg,P<0.01),respectively.Acupuncture-treatment,significantly raise the effect of SOD、GSH-PX,attenuated the expression of MDA.Conclusions In 6-OHDA lesion striatum model,acupuncture-treatment significantly elevated the effect of SOD & GSH-px,attenuated the expression of MDA.Acupuncture may take part in adjust of mechanism of oxidative stress in Parkinson’s disease.
