【摘要】 目的观察淫羊藿甙对关节磨屑刺激单核细胞分泌IL-1β、IL-6、PGE2、TNF的影响。方法分离培养兔外周血单核细胞,分成4组。D组为关节磨屑组:培养细胞中加入体积比为0.1%的Ti微粒。A、B、C组分别在D组的基础上加入10mg/mL、1mg/mL、0.1mg/mL的淫羊藿甙。用放射免疫测定法测量各上清液中PGE2及TNF含量;用酶联免疫吸附试验测定各上清中IL-1β及IL-6含量。结果与D组比较,A、B、C三组各检测指标均明显下降(P<0.01),且与药物浓度呈负相关。结论淫羊藿甙可以降低假体周围炎性细胞因子的含量,从而防治假体松动的产生。更多还原

【Abstract】 Objective To study the effect of Icariin on IL-1β、IL-6、PGE2、TNF expression of human monocytes with wear particles stimulation.Methods In vitro rabbit peripheral blood mononuclear cells(PBMC) were cultured in 4 groups.Group D(underwent wear particles stimulation by adding Ti particles of 0.1% volume ratio during culting);Group A(D plus 10mg/ml Icariin);Group B(D plus 1mg/ml Icariin);Group C(D plus 0.1 mg/ml Icariin).PGE2 and TNF contents of supernatant were measured by radioimmunoassay.ELISA was applied for determination of contents of IL-1β and IL-6.Results Compared with Group D,Groups A,B and C appeared obvious decline in observed indexes,with negative correlation.Conclusion Icariin is able to prevent prosthetic loosening by lowering contents of inflammatory factors around prosthesis.更多还原
