黄海葵高效液相色谱指纹图谱研究

【摘要】 目的应用高效液相色谱法(HPLC)建立黄海葵的HPLC指纹图谱,为其质量控制和药材鉴别提供依据。方法采用反相C18色谱柱,以乙腈-0.1%(V/V)磷酸水为流动相,二元线性梯度洗脱,流速为1.0 mL.min-1,柱温为室温,检测波长为210 nm,进样量5μL;采用高效液相色谱-质谱联用技术(HPLC-MS)对其特征峰进行快速分析;将相似度计算法用于8批黄海葵药材的评价。结果以7个共有峰为评价指标,建立了黄海葵药材的HPLC指纹图谱,通过相似度计算发现8批黄海葵相似度均在0.97以上;HPLC-MS分析获得的4个特征峰的质谱信息,可实现黄海葵药材指纹图谱特征峰的准确定位。结论该方法准确、简单、稳定性好,所建立的黄海葵药材的HPLC指纹图谱可作为药材定性真伪鉴别和质量控制的有效手段。

【Abstract】 Objective To establish a high performance liquid chromatography(HPLC)fingerprinting method for the quality evaluation and discrimination of Anthopleura xanthogrammica(Berkly).Methods The chromatographic fingerprints of the 8 samples were determined by injecting 5 μL of the sample solution each time on a reversed phase C18column with the gradient elution solvent system composed of water and acetonitrile.The flow rate was 1.0 mL·min-1,the column temperature was maintained at room temperature and the signals were acquired at 210 nm.Characteristic peaks of the fingerprint could be rapidly analyzed by high performance liquid chromatography-mass spectrometry(HPLC-MS),and the method of similarity analysis was used to evaluate the 8 samples.Results The HPLC fingerprint of Anthopleura xanthogrammica(Berkly) was set up,which included 7 common peaks,and the similarities of 8 samples were all more than 0.97.Mass spectra of the 4 characteristic peaks obtained using HPLC-MS could be used to accurately confirm the characteristic and common peaks in fingerprint of Anthopleura xanthogrammica(Berkly).Conclusion The method is stable,simple and accurate,and could be used for the identification and quality control of Anthopleura xanthogrammica(Berkly).
