资木瓜皂苷提取工艺的优化及对大鼠佐剂性关节炎抗炎免疫机制的研究

【摘要】 目的研究资木瓜皂苷(SCS)提取工艺的优化及对大鼠佐剂性关节炎(AA)的抗炎作用及免疫机制。方法以SCS的提取总量为考察指标,采取正交试验法考察乙醇浓度、溶媒用量、提取时间、提取次数等因素,每个因素设置3个水平。流式细胞术检测SCS对AA外周血中CD4+、CD8+的表达水平;MTT法检测SCS对AA外周T淋巴细胞增殖的影响。结果 SCS最佳提取工艺为乙醇浓度70%,溶媒容量10倍,提取时间1h,提取3次,提取率为1.7%。SCS能降低AA外周血中CD4+的表达水平,对CD8+的表达无明显影响,对AA外周T淋巴细胞增殖有抑制作用。结论最佳提取条件为A2B1C2D3,SCS通过抑制T淋巴细胞增殖和降低CD4+表达而发挥抗炎免疫作用。

【Abstract】 OBJECTIVE To study optimization on extraction technology of saponin of Chaenomeles Speciosa( SCS) and study on its anti-inflammatory immunologic mechanism to rats adjuvant arthritis( AA) . METHODS Orthogonal design L9 ( 34 ) on four factors including alcohol level,alcohol volume,extraction time and extraction frequency was used,and took the saponin rate as the index. MTT was used to determine lymphocytes proliferative activity. T lymphocytes subgroup CD4 + and CD8 + in peripheral blood were detected by flow cytometry. RESULTS The best technology was at alcohol level 70% ,10-fold alcohol volum,extracting 1 h and 3 times,saponin yield from chaenomeles was 1. 7% . SCS decreased CD4 + expression in peripheral blood,but SCS did not show the ability to advance the level of CD8 + ,SCS also had effect on inhibiting T lymphocyte proliferation. CONCLUSION The best technology was A2 B1 C2 D3 . The anti-inflammatory immunologic mechanism of SCS includes supressing T lymphocyte proliferation and decreasing CD4 + expression.
