荞麦花叶中芦丁对血管紧张素Ⅱ诱导的新生大鼠心肌细胞肥大的体外抑制作用
In vitro inhibitory effects of rutin from buckwheat flowers and leaves on angiotensin II induced cardiomyocytes hypertrophy of neonatal rat
【摘要】 目的观察荞麦花叶中芦丁(RBFL)对新生大鼠心肌细胞肥大的作用,并探讨其对蛋白激酶C(PKC)信号途径的影响。方法选用培养的新生大鼠心肌细胞为研究对象,以10-7mol·L-1血管紧张素Ⅱ(AngⅡ)刺激建立体外心肌细胞肥大模型,并用不同浓度的RBFL作用于细胞,测定心肌细胞表面积,3H-亮氨酸掺入法测定心肌细胞蛋白质的合成速率,免疫细胞化学法测定心肌细胞PKC蛋白的表达,同位素底物掺入法检测PKC活性。结果 10-7mol·L-1AngⅡ能显著增加心肌细胞表面积和蛋白质合成速率、PKC蛋白的表达及其活性;1~10mg·L-1RBFL能剂量依赖性地抑制AngⅡ诱导的心肌细胞表面积和蛋白质合成速率的增加、PKC蛋白的表达及其活性,并具有统计学意义。结论 RBFL能明显抑制培养的新生大鼠心肌细胞的肥大,此作用可能与其对心肌细胞中PKC信号途径的调控有关。
【Abstract】 OBJECTIVE To observe the effects of rutin from buckwheat flowers and leaves(RBFL) on angiotensin II-induced cardiomyocytes hypertrophy and on protein kinase C(PKC) signaling pathway.METHODS Cardiomyocytes hypertrophy model was made by adding 10-7 mol·L-1 angiotensin II(AngⅡ) to the medium of cultured cardiomyocytes of neonatal rats,meanwhile,different concentrations of RBFL were applied to observe their effects.Cardiomyocytes surface area was analyzed and protein synthesis rate was measured by 3H-Leucine incorporation as the index of cardiomyocytes hypertrophy.The expression of PKC protein in cardiomyocytes was observed with immunocytochemistry method,and the activity of PKC in cardiomyocytes was assayed by incubating PKC with histone III S and γ-32P ATP.RESULTS 10-7 mol·L-1 AngⅡ could remarkably increase cell surface area and the protein synthesis rate,the expression of PKC protein and the activity of PKC in cultured cardiomyocytes of neonatal rats,which could be inhibited by 1-10 mg·L-1 RBFL dose-dependent and the results had statistical significance.CONCLUSION RBFL had obvious inhibitory effect on angiotensin II-induced cardiomyocytes hypertrophy,which might be due to the regulation on PKC signaling pathway.
