Basigin mRNA在早期缺血心肌和非缺血心肌的表达变化

【摘要】 目的探讨Basigin mRNA在早期心肌缺血大鼠模型缺血心肌(EI M)和非缺血心肌(NI M)中的表达及意义。方法以β-actin为内对照,应用real-ti me PCR方法检测0 min、15 min、30 min、60 min早期缺血心肌和非缺血心肌Basigin mRNA的表达情况。结果EI M、NI M、假手术(SO)组心肌和非手术组心肌Basigin mRNA表达在0 min差异没有统计学意义(P>0.05)。EI M组在心肌缺血15 min、30 min和60 min较0 min时Basigin mRNA表达降低,差异有统计学意义(P<0.001),NI M组和SO组在不同时间段之间差异无统计学意义(P>0.05);EI M组和NI M组在30 min、60 min时Basigin mRNA表达的差异有统计学意义(P<0.05)。结论Basigin在缺血30min参与心肌的病理生理过程,Basigin在鉴定早期心肌缺血死亡的案例中有十分重要的法医学意义。

【Abstract】 Objective To investigate the expression of Basigin mRNA in early ischemic myocardium(EIM) and non-ischemic myocardium(NIM) in rat.Methods Real-time polymerase chain reaction(PCR) technique was applied for detecting Basigin mRNA andβ-actin expression in EIM and NIM at 0 min,15 min,30 min,and 60 min.Results No significant differences of the Basigin mRNA expression in EIM and NIM between 0 min and sham operation(SO) or non-operation group(P>0.05) were observed.Basigin mRNA expression in EIM significantly decreased in 15 min,30 min,and 60 min group compared with that of 0 min group,respectively(P<0.001).However,we failed to find any significant difference in NIM and NO from 0 min to 60 min.Basigin mRNA expression in EIM decreased significantly compared with that of in NIM(P<0.05) after myocardial ischemia for 30 min.Conclusion Basigin was involved in myocardium pathophysiology process after myocardial ischemia for 30 min,indicating that Basigin may be identified as a predictor of early myocardial ischemia in forensic medicine.
