野菊花中绿原酸的不同提取工艺比较

【摘要】 目的:以绿原酸含量和浸膏率为考察指标,探讨从野菊花中提取该成分的新工艺。方法:采用加热回流提取法、组织破碎提取法及超声波提取法3种提取工艺,利用反相高效液相色谱法,以甲醇:冰醋酸:水(体积比为18:1:81)为流动相(流速为1.0mL/min),测定3种提取方法的绿原酸含量及浸膏率。结果:绿原酸在0.01518~0.15180g/L范围内线性关系良好(r=0.9999)。加热回流提取法绿原酸含量为(0.4211±0.0137)%,超声波提取法为(0.3361±0.0107)%,组织破碎提取法为(0.4066±0.0065)%,3者相比差异有统计学意义(F=28.061,P=0.001)。加热回流提取法的浸膏率为(32.48±0.68)%,超声波提取法为(22.53±1.16)%,组织破碎提取法为(26.79±0.55)%,3者相比差异有统计学意义(F=52.413,P<0.001)。结论:组织破碎提取法具有简便迅速和节能高效的优点,是一种值得推广的提取新工艺。

【Abstract】 Aim:To study the effect of different extraction methods on the content of chlorogenic acid and extraction from Flos Chrysanthemi Indici.Methods:To extract Flos Chrysanthemi Indici through three different extraction methods. The content of chlorogenic acid in Flos Chrysanthemi Indici was quantitated by using RP-HPLC on the Agilent XDE C18 (150 mm×4.6 mm,5 μm) column with the mobile phase consisted of methanol,acetic acid,water (18:1:81),flow rate:1.0mL/min.Results:The linear ranges for chlorogenic acid was 0.0151 8~0.151 80 g/L(r=0.999 9).The content of chlorogenic acid exacted by heating and refluxing was (0.421 1±0.013 7)%,and exacted by ultrasonic extraction was (0.336 1±0.010 7)%,exacted by tissue-smashing exaction was (0.406 6±0.006 5)%(F=28.061,P=0.001).The content of extraction extracted by heating and refluxing was(32.48±0.68)%,and exacted by ultrasonic extraction was(22.53±1.16)%,exacted by tissue-smashing extraction was(26.79±0.55)%(F=52.413,P<0.001).Conclusion:The methods of tissue-smashing extraction is a new extraction method that is worth to popularizing for simpliness and quickness and high efficiency.
