醋酸铅对大鼠睾丸支持细胞活性及抑制素B mRNA水平的影响

【摘要】 目的:观察醋酸铅对体外培养大鼠睾丸支持细胞活性和抑制素B(INHB)mRNA水平的影响。方法:建立大鼠睾丸支持细胞体外培养模型,分别给予不同浓度(0、10-8、10-7、10-6和10-5mol/L)的醋酸铅处理细胞,培养24h后采用MTT法测定细胞活性;同法分组,采用RT-PCR检测INH B mRNA的表达。结果:各组细胞活性比较,差异无统计学意义(F=2.451,P=0.064);各组细胞INH B mRNA的相对表达量比较,差异有统计学意义(F=9984.370,P<0.001),醋酸铅浓度在10-8和10-7mol/L时,睾丸支持细胞细胞INH B mRNA的表达水平低于对照组(P<0.05)。结论:醋酸铅对大鼠睾丸支持细胞可能有毒性作用。

【Abstract】 Aim:To investigate the effects of lead acetate on the cell activity and the expression level of inhibin B(INH B) mRNA in Sertoli cells of rats.Methods:The Sertoli cells were allocated into 5 groups and given lead acetate at 0,10-8,10-7,10-6,and 10-5mol/L,respectively.MTT method was used to observe the cell activity.The Sertoli cells were allocated as above,and the INH B mRNA level was measured by RT-PCR.Results:The cell activity of Sertoli cells had no significant difference among the 5 groups(F=2.451,P=0.064). The expression level of INH B mRNA had significant difference among the 5 groups(F=9 984.370,P<0.001),and the expression levels of INH B mRNA in 10-8 and 10-7mol/L groups were higher than that of control group(P<0.05).Conclusion:Lead acetate could affect the expression level of INH B mRNA in Sertoli cells,which has toxic effects on the Sertoli cells.
