钙离子胁迫诱导大豆根尖细胞凋亡
APOPTOSIS INDUCED BY Ca2+ IRON STRESS ON ROOT TIP CELLS OF SOYBEAN
【摘要】 用生长中的大豆根尖细胞为材料,经不同浓度CaCl2和不同时间处理,通过对凋亡细胞的形态观察和基因组DNA分析,探究CaCl2处理浓度和时间对根尖细胞凋亡影响。结果表明:利用CaCl2溶液作诱导剂处理生长的大豆根尖细胞,能引起细胞凋亡。在凋亡过程中,凋亡细胞的形态结构发生变化,DNA降解成大小不等的片段;细胞凋亡率与诱导剂处理浓度和时间正相关;在处理浓度为0.2mol/L和时间为12h时,凋亡率接近50%。因此,降低大豆根尖细胞凋亡率CaC12胁迫浓度应<0.10mol/L。
【Abstract】 CaCl2 was used as an inducing agent to treat growing soybean root tip with different concentrations and time.The apoptosis of root tip cells was studied via analyzing the variations of morphological characteristics and genomic DNA.The results showed that the treatment with CaCl2 could lead to root tip cell apoptosis.Apoptosis cell morphology changed during apoptosis and genomic DNA degradated into fragments of various sizes.The apoptosis rate was positively related to the treated concentration and time.The apoptosis rate nearly reached 50% under the treatment 0.2mol /L CaCl2 and 12 hours.But apoptosis rate declined when the concentration of CaCl2 was lower than 0.10mol /L.
