拜阿司匹灵预处理对高血糖SD大鼠局部脑缺血再灌注损伤中ET-1的表达

【摘要】 目的采用高血糖条件下SD大鼠局灶性脑缺血再灌注损伤模型,通过测量大鼠脑梗死体积及内皮素-1(Endo-thelin-1,ET-1)的表达情况,探讨预防性应用拜阿司匹灵对高血糖条件下SD大鼠局灶性脑缺血再灌注损伤的脑保护作用。方法随机分为对照组与拜阿司匹灵组,2组按缺血90 min再灌注3 h、6 h1、2 h再分为3个亚组。2组均建立高血糖模型及SD大鼠右侧大脑中动脉缺血再灌注模型。病理图像分析仪测量脑梗死体积,免疫组化方法测定ET-1的表达情况。结果拜阿司匹灵组与对照组相同再灌注时间点相比较:梗死体积减小(P均<0.01);缺血区表达ET-1减少(P均<0.05)。结论预防性应用拜阿司匹灵能减轻高血糖条件下的局灶性脑缺血再灌注损伤,缩小梗死体积,使损伤脑区ET-1表达减少。

【Abstract】 Objective To discuss protective effect of pretreatment of bay aspirin on the condition of hyperglycemia by focal ischemia-reperfusion model of SD rat on the condition of hyperglycemia,through cerebral infarction volume and expression of Endothelin-1.Methods These patients were randomly divided into control group and bay aspirin group,each group was divided into 3 subgroups according to reperfusion 3 h,6 h and 24 h after ischemia for 90 minutes.Hyperglycemia model and ischemia-reperfusion model in right cerebral middle artery of SD rat were established.The infarction volume with the pathology image analysis system measured and the expression of ET-1 with immunohistological method was observed.Results omparison between bay aspirin group and hyperglycemia group at the same point of time: infarction volume decreased,P<0.01,and ET-1 at the ischemia area decreased,P<0.05.Conclusion Under condition of hyperglycemia,preventive pretreatment of bay aspirin could decrease focal ischemia-reperfusion damage,reduce infarcted volume,reduce the expression of ET-1.
