猪α干扰素的表达及在PAM细胞系中抗PRRSV活性检测

【摘要】 为研究猪α干扰素蛋白功能和了解猪α干扰素蛋白的抗病毒活性,扩增并测序了猪α干扰素基因(IFN-α),并克隆至原核表达载体pET-28a中,构建原核表达载体pET-IFN-α,将pET-IFN-α转化至BL21进行表达。Western-Blot结果表明,表达的蛋白具有很好的生物学活性。用细胞病变抑制法在PAM细胞系上进行抗病毒活性测定,结果表明,猪α干扰素有较为显著的抗PRRSV活性,其抗病毒活性效价约为1.13×106U/mg。

【Abstract】 To study the function of porcine interferon-α (PoIFN-α),the gene of PoIFN-α was cloned and inserted into the prokaryotic expression vector pET-28a to construct the recombinant plasmid pET-IFN-α,and then transformed into E.coli BL21.The recombinant protein was purified and renatured from inclusion bodies to obtain a native state of biological activity.Antiviral activity assays for porcine interferon were performed and evaluated by standard procedure in the cell/virus test system of PAM/PRRSV.The results showed that PoIFN-α demonstrated significant antiviral activity,which is 1.13×10 6 U/mg.
