辣椒细胞质雄性不育系及其保持系的AFLP分析(英文)

【摘要】 [目的]对辣椒细胞质雄性不育系及其保持系进行AFLP分析,为进一步深入研究辣椒细胞质雄性不育的分子机制奠定基础。[方法]采用AFLP技术对辣椒细胞质雄性不育系21A及其保持系21B进行分析,获得不育系21A特异扩增片段。将特异扩增片段回收测序并用BLASTn和BLASTX程序在NCBI数据库中进行同源性检索和相似性比对。[结果]获得了不育系21A特异性扩增片段AA/CAG169,该片段中部分序列(位于122~158bp,AGAATTGGTA CGCAGTCTATGATGAGTCCTGAGTAAC,共37bp)是Penaeus monodon性别及生长性状相关的AFLP标记序列(AFE549M27.1)中串联重复序列;BLASTX和TBLASTX分析结果表明,该特异性片段的翻译产物与马铃薯的假拟gag-pol蛋白、假拟逆转座子蛋白、假拟逆转座子gag蛋白具有较高的相似性。[结论]特异性片段AA/CAG169的功能可能与逆转座子有关,表明其可能与育性密切相关。

【Abstract】 [Objective] The aim of this study was to analyze the cytoplasmic male sterile line 21A and its maintainer line 21B of peppers by AFLP,and lay the foundation for further studies on molecular mechanism of the cytoplasmic male sterility in peppers.[Method] Cytoplasmic male sterility(CMS)line 21A and its maintainer line 21B were analyzed by AFLP to obtain the specific amplified fragments of cytoplasmic male sterile line 21A,while the specific amplified fragments were recovered or sequenced,and analyzed by BLASTn and TBLASTX in GenBank of NCBI.[Result] A specific fragment AA/CAG169 was obtained from cytoplamic male sterility(CMS)line 21A.Partial DNA sequence of AA/CAG169 was the series repeated sequence in AFLP marker sequence related to the sex and growth traits of Penaeus monodon.The results of BLASTX and TBLASTX showed that the translation products of this specific fragment had a higher similarity to Solanum demissum putative gag-pol protein,Solanum demissum putative retrotransposon protein and Solanum demissum putative retrotransposon gag protein respectively.[Conclusion] The function of this specific fragement AA/CAG169 is more likely to be closely related to retrotransposon,which indicates that the special fragment is possibly related to the sterility trait.
