亨廷顿蛋白相关蛋白1在胚胎期大鼠脑内的表达

【摘要】 目的观察亨廷顿蛋白相关蛋白1(huntingtin-associated protein 1,HAP1)在胚胎不同时期SD大鼠脑内的表达。方法采用免疫组织化学方法,观察胚胎期SD大鼠不同脑区HAP1的表达情况。结果 E6.5组胚胎中未发现HAP1的表达;在E8.5组,神经上皮可见一定的HAP1的表达及stigmoid小体;在E11.5组胚胎的脑组织中HAP1的表达略有增加,阳性细胞多呈中小型圆形,少见阳性纤维;在E14.5组,HAP1的强阳性信号出现在大脑皮质、后脑、末脑,丘脑、下丘脑、海马及纹状体的表达稍弱,细胞小而圆,排列密集;在E17.5组,HAP1的强表达主要集中在丘脑、下丘脑、脑桥、延髓、杏仁海马区和齿状回,并可见密集深染的阳性纤维;P0组和E17.5组比,HAP1的表达呈下降趋势。stigmoid小体数目的变化趋势与HAP1基本一致。结论 HAP1在胚胎早期即有表达,并逐渐增多,中期明显增加并持续到出生,提示HAP1可能参与中枢神经系统的形成和成熟机制。

【Abstract】 Objective To study the expression of huntingtin - associated protein 1 ( HAP1) in embryonic encephalon of SD rats at different stages.Methods With immunohistochemical method,the expression of HAP1 in different areas of embryonic encephalon of SD rats was observed.Results In Croup E6.5,no HAP1 expression was found.In E8.5,some expression and stigmoid bodies was observed in neuroepithelium.In E11.5,the expression was increased slightly,the positive cells were mainly medium - small - sized round and positive fibers were few.In E14.5,strongly positive signal of HAP1 appeared in cerebral cortex,metencephalon,myelencephalon,thalamus,hypothalamus.The signals in hippocampus and corpus striatum were sligntly weaker than that of the former.The cells were small, round,and arranged closely.In E17.5,strong expression was found mainly in thalamus、hypothalamus、 pons、medulla oblongata,amygdalohippocampal area and dentate gyrus,and there were coarctated deeply -stained positive fibers.Compared with E17.5,the expression of P0 decreased.Tendency of the change of stigmoid bodies correspond to the expression of HAP1.Conclusion The expression of HAP1 begins at early embryonic stage,and gradually increases.From middle embryonic stage to birth,the expression increases significantly,which hint that HAP1 may be related to the forming and development of central nervous system.
