一氧化碳对硫化氢舒张大鼠肺动脉作用的影响
THE EFFECTS OF CARBON MONOXIDE ON THE RAT PULMONARY ARTERY RELAXATION RESPONSES TO HYDROGEN SULFIDE
【摘要】 目的观察在脂多糖(lipopolysaccharide,LPS)诱导下,一氧化碳(carbon monoxide,CO)对硫化氢(hydrogen sulfide,H2S)舒张肺动脉作用的影响。方法气管内滴注生理盐水或LPS后,制备肺动脉环(pulmonaryartery rings,PARs),应用血管环张力测定技术,分别在给予CO供体氯血红素(hemin,Hm)或血红素氧合酶1抑制剂锌原卟啉9(zinc protoporphyrin-IX,ZnPP-IX)的条件下,从离体水平观察PARs对H2S供体硫氢化钠(sodiumhydrosulfide,NaHS)舒张反应变化,同时检测出肺血(out-going pulmonary blood,OPB)和入肺血(in-flowing pulmonaryblood,IPB)中碳氧血红蛋白(carboxyhemoglobin,COHb)含量,以其差值反映肺循环CO生成的水平。结果滴注LPS4 h和8 h后,用Hm孵育PARs后,PARs对NaHS的累积浓度舒张反应较孵育前显著增强(P<0.01),曲线左移;相反在滴注LPS 8 h后,用ZnPP-IX孵育PARs,PARs对NaHS的累积浓度舒张反应较孵育前明显减弱(P<0.01),曲线右移,而在滴注4h后,ZnPP-IX对PARs的舒张反应无明显影响。与对照组相比,滴注LPS 8 h后,OPB与IPB中COHb含量的差值明显增高(P<0.05),而滴注LPS 4 h后无明显变化(P>0.05)。结论在LPS诱导下,CO可以增强H2S舒张肺动脉的作用。
【Abstract】 Objective To explore the effect of carbon monoxide on the rat pulmonary artery relaxation responses to hydrogen sulfide. Methods After NS or lipopolysaccharide (LPS) instillation intratracheally, the pulmonary artery rings (PARs) were prepared and incubated with a donor of CO hemin(Hm) or an inhibitor of heme oxygenase-1 Zinc protoporphyrin-Ⅸ(ZnPP-Ⅸ),then the changes of cumulative relaxation response of pulmonary artery rings ( PARs) to sodium hydrosulfide ( NaHS) were detected in vitro. In addition, the contents of carboxyheraoglobin (COHb) in out-going pulmonary blood (OPB) and in-flowing pulmonary blood(IPB) were measured,and the difference between the contents of COHb in OPB and that of IPB was calculated to represent CO content. Results Four and eight hours after instilling LPS,cumulative relaxation response of PARs to NaHS was enhanced by incubation with Hm,and the curve shifted left. The contrary effect was observed by incubation with ZnPP-Ⅸ 8 h after instilling LPS,but no significant change was shown 4 h after instilling LPS. The difference between the contents of COHb in OPB and that of IPB increased 8 h after instilling LPS,but no significant change was shown 4 h after instilling LPS. Conclusion The production of CO induced by LPS can enhance the rat pulmonary artery relaxation responses to hydrogen sulfide.
