两种滑刃亚目线虫的ITS-PCR-RFLP研究

【摘要】 采用ITS区通用引物对阿苏里伞滑刃线虫(Bursaphelenchus arthuri)和燕麦滑刃线虫(Aphelenchusavenae)的基因组DNA进行PCR扩增,对其PCR扩增产物进行序列测定分析,同时用5种限制性内切酶(RsaⅠ、HaeⅢ、MspⅠ、HinfⅠ和AluⅠ)对PCR扩增产物进行酶切,得到相应酶切图谱。两种线虫的DNA序列测定分析以及ITS-RFLP酶切图谱得到的鉴定结果与形态鉴定结果一致。试验得到的阿苏里伞滑刃线虫和燕麦滑刃线虫ITS-RFLP酶切图谱可以为线虫的分子鉴定提供参考。

【Abstract】 In this paper,the universal primers was used for expansion ITS district of Bursaphelenchus arthuri and Aphelenchus avenae from their genomic DNA.The sequence of PCR products was analysed and the five restriction enzymes(Rsa Ⅰ,HaeⅢ,MspⅠ,HinfⅠ and AluⅠ) were tested to cut the PCR products in order to acquire the restriction enzyme mapping.The results of ITS sequence analysis and ITS-PCR-RFLP were consistent with the morphological identification for B.arthuri and A.avenae.The established restriction enzyme mapping of B.arthuri and A.avenae would provide a reference for the nematode molecular identification in the future.
