运用均匀设计方法优化蓖麻变应原提取工艺
Extraction Process of Allergens in Castor Bean Meal by Uniform Design
【摘要】 以均匀设计法设计提取蓖麻变应原的实验条件,探讨了影响蓖麻饼粕中蓖麻变应原乙醇提取率的因素.研究了乙醇浓度、提取时间、提取温度及料液比等因素对蓖麻变应原提取率的影响,得到了最佳优化工艺条件.用SDS-PAGE凝胶电泳检测蓖麻变应原的纯度,紫外扫描、高效液相检测、红外光谱分析蓖麻变应原的结构,提取物以蓖麻变应原的光吸收值为指标进行检测.优选出从蓖麻饼粕中提取蓖麻变应原的最佳工艺为:乙醇浓度为85%,提取时间为8 h,提取温度为50℃,提取物料比为1∶21(W/V).用该工艺提取出的蓖麻变应原,经高效液相检测纯度达到了98%.
【Abstract】 The extraction condition of allergens from castor bean by uniform design were researched.Not only the influences of reaction ethanol concentration,temperature,time,and raw materials ratio on process,but also the best condition was obtained.SDS-PAGE was used to detect the purification of the allergen,and UV scanning,HPLC,and IR spectroscopy were used to characterize its structure and distribution.The results show that the optimal extraction condition was 85% ethanol concentration,8 h extraction time at 50 ℃ and the raw materials ratio of 1∶21(W/V).The purity of the experiment can reach 98%.
