复合前驱体溶胶—凝胶法固定木瓜蛋白酶条件的优化

【摘要】 以正硅酸甲酯(TMOS)和甲基三甲氧基硅烷(MTMS)为复合前驱体,以聚乙二醇400(PEG400)为稳定剂,采用溶胶—凝胶法固定木瓜蛋白酶.利用单因素和正交试验设计,得到最佳的固定化条件为:MTMS与TMOS的体积比4∶1,水与硅源的体积比(R值)0.3,用5mL前驱体水解液制备成凝胶,0.1mol/L盐酸溶液的用量210L,PEG400的用量为240L,给酶量150L.在优化条件下制得的固定化酶包封率为84.1%,活力回收率为84.4%.

【Abstract】 Papain was immobilized in silica particles by a sol-gel process, in which methyl silicate (TMOS) and meth-yltrimethoxylsilane (MTMS) were used as precursors. The immobilization conditions were optimized: MTMS and TMOS volume ratio 4∶1, water and silica source volume ratio (R value) 0.3, the gel was prepared by 5 mL precursor mixture, hydrochloric acid solution (0.1 mol/L) 210 L, PEG400 240 L, the amount of enzyme solution 150 L. The entrapment efficiency of papain prepared under the optimal conditions was 84.1 %, and the activity recovery rate was 84.4 %.
