扩张力作用下大鼠矢状缝形态学改变与ALP活性关系初探

【摘要】 目的分析扩张力作用下大鼠矢状缝形态改变与成骨细胞活性的关系。方法选用鼠龄7周左右的SD大鼠72只,随机分到正常对照组、手术对照组、实验加力组,分别在实验第4天、第7天和第14天时各处死8只。取材制片后,计算骨缝组织中成骨细胞ALP染色阳性表达率,并通过显微图像分析系统测量骨缝组织中骨-骨缝边缘长、骨缝组织面积,对扩张力作用下骨缝改建过程中骨缝组织形态与成骨细胞活性的关系进行分析。采用SPSS11.0软件包对数据进行单因素方差分析。结果 (1)实验加力组在100g扩张力作用下,第4天组和第7天组实验动物的骨缝组织面积显著增加。(2)加力后第7天,实验加力组动物的骨缝组织中骨-骨缝边缘长、面积和成骨细胞ALP阳性表达率最高。(3)加力后第14天,实验加力组动物的骨缝组织中骨-骨缝边缘长与加力后第7天组比较差异无统计学意义。结论 (1)100g扩张力作用下,持续到加力后第7天骨缝纤维组织保持活跃增殖。(2)第7天时骨缝组织中ALP活性最高,成骨细胞功能最活跃。(3)第14天时沿着骨缝边缘的骨沉积和骨基质矿化持续活跃进行。

【Abstract】 Objective To analyze the relationship between suture’s morphology and ALP’s activity in the suture model.Methods 72 Sprague-Dawley rats were divided into a baseline group,a control group and an experimental group at random,and were sacrificed 4,7 and 14 days after application of the initial tensile force of 100g.The suture sections were stained with hematoxylin eosin to observe the general morphological changes of the suture,and the parameters of bone-suture length and suture area were valued by TIGER image analyzer.Osteoblasts were tested by alkaline phosphatase using cytochemical method and the number of positive stained osteoblasts was counted to calculate the percent of positive cell along the edge of suture.The data of the three parameter were analyzed with SPSS 11.0 software package for one-way ANOVA.Results The suture area in the experimental group increased significantly at the time points of 4th day and 7th day under 100g tensile force.The level of bone-suture length and the percent of active stained osteoblasts in the experimental group reached the highest at the time point of 7th day.The suture area was similar at the two time points of 4th day and 7th day in the experimental group.Conclusion The connective tissue of the interparietal suture appeared in the state of continuously overwhelming fibrogenesis until 7 days of 100g tensile force application.The activity of osteogenesis reached the highest at the time point of 7th day.The bone matrix formation and mineralization remained active at the time point of 14th day.
